摘要
目的研究H_2O_2对SD大鼠海马-氨基-3-羟基-5-甲基-4-异恶唑丙酸(AMPA)受体棕榈酰化水平的影响。方法用400 M H_2O_2孵育正常大鼠急性脑片后,分离海马组织;酰基生物素交换技术提取AMPA受体亚基GluA1、GluA2棕榈酰化蛋白;蛋白免疫印迹技术检测上述蛋白表达水平。结果与对照组相比,H_2O_2升高大鼠海马蛋白棕榈酰化总水平、Glu A1(P<0.05)和Glu A2(P<0.01)棕榈酰化水平。结论400 MH_2O_2可显著提高SD大鼠海马AMPA受体棕榈酰化水平。
Objective To investigate the effects on palmitoylated modification of AMPA receptors in hippocampus of rats by hydrogen peroxide (H2O2 ). Methods The palmitoylated protein ofGluA1, GluA2 in the hippocampus of the acute slices for rats after incubated 400μM H2O2 were extracted through acyl-biotinyl exchange method. The expression levels of extracted protein were detected by western blotting. Results Compared with the control rats, the global palmitoylated level in the hip- pocampus of the incubated H2O2 rats was significantly increased, as well as the palmitoylation of GluA1 (P〈0.05) and GluA2 (P〈0.01) in hippocampus of the incubated H2O2 rats were remarkably elevated. Conclusion The present study indicated that the palmitoylated levels of AMPA receptors in hippocampus of rats were increased by 400μ M H2O2.
