藤芪蚤复方的提取及其体外抗肿瘤活性研究

摘要 目的:探究藤芪蚤复方的醇提工艺及其提取物体外抗肿瘤活性。方法:以总黄酮含量为考察指标,采用正交试验方法对藤芪蚤中药复方醇提工艺条件优化;并采用CCK-8法评价藤芪蚤中药复方醇提物的体外抗癌活性。结果:藤芪蚤中药复方优选的提取工艺:用12倍和10倍80%乙醇回流提取2次,提取时间为1.0 h;藤芪蚤中药复方醇提取物对人肺癌A549细胞、人骨肉瘤143B细胞、人鼻咽癌CNE-2Z细胞及人急性早幼粒细胞白血病HL60细胞都有明显的体外抗癌活性,其最大抑制率与CC50值(总黄酮的浓度)分别为:99.43%、98.60%、99.21%、74.81%和156.75μg/m L、61.42μg/m L、143.94μg/m L、232.49μg/m L。结论:中药复方醇提工艺简单可行,其提取物对肺癌细胞、鼻咽癌细胞、骨肉瘤细胞及急性早幼粒细胞白血病细胞均具有明显的抑制作用,提示藤芪蚤复方醇提物具抗肿瘤作用,可作为进一步开发参考。 Objective: To study the Ethanol extraction technology and its antitumor activity in vitro. Methods: The total flavonoid content was used as an index to study the optimum conditions of the extraction of Ethanol from the traditional Chinese medicine. The CCK-8 method was used to evaluate the anti-cancer activity of the compound extract of the traditional Chinese medicine. Results: The extraction process was the best extraction process of Tengqizao traditional Chinese medicine compound. The extraction time was 1.0 h with 12 times and 10 times 80% ethanol, and the extraction time was 1.0 h. The extract of Tengqizao flea compound on human lung cancer A549 cells, human osteosarcoma 143B cells The highest inhibitory rate and CC50 value (total flavonoid concentration) of human nasopharyngeal carcinoma CNE-2Z cells and human acute promyelocytic leukemia HL60 cells were 99.43 % and 98.60 % respectively, 99.21%, 74.81% and 156.75 μg/mL, 61.42 μg/mL, 143.94 μg/mL, 232.49 μg/mL. Conelusion:The traditional Chinese medicine compound alcohol extraction process is simple and feasible, its extract on hmg cancer cells, nasopharyngeal carcinoma cells, osteosarcoma cells and acute promyelocytic leukemia cells have a signilicant inhibitory effect, suggesting that there is a certain anti- Tumor effect.
