摘要
目的观察高糖对大鼠膀胱平滑肌细胞(BSMCs)中钙库操控的钙离子通道(SOCC)的影响。方法通过激光共聚焦显微镜观察记录高糖处理的BSMCs激动和抑制SOCC后细胞内相对荧光强度值,反映其钙离子浓度变化。通过RT-qPCR和Western blot检测SOCC相关组成分子STIM1和Orai1在高糖处理的BSMCs中mRNA与蛋白的表达水平。结果高糖处理的BSMCs加入SOCC激动剂CPA后相对荧光强度显著增加,加入SOCC抑制剂SKF-96365后相对荧光强度显著降低。高糖能上调BSMCs中SOCC重要组成分子STIM1和Orai1的mRNA表达水平以及蛋白表达水平。结论高糖能促进大鼠BSMCs中SOCC的激活,使胞外Ca^(2+)内流,可能为糖尿病膀胱的病因学研究提供新的思路。
Objective To explore the effect of high glucose on store-operated calcium channel (SOCC) of rat bladder smooth muscle cells (BSMCs). Methods After rat BSMCs were treated with high glucose,the relative fluorescence intensity of cells were detected with laser scanning confocal microscope to reflect calcium ion concentration. The expressions of stromal interaction molecule1 (STIM1) and calcium release-activated calcium channel protein 1 (Orail) were detected with quantitative real-time polymerase chain reaction (RT-qPCR) and Western blotting. Results Confocal microscope results showed that the relative fluorescence intensity of BSMCs treated with high glucose increased significantly after treatment of SOCC agonist CPA and declined significantly after treatment of SOCC inhibitor SKF-96365. The expressions of STIM1 and Orail could be elevated at both mRNA and protein levels by high glucose. Conclusion High glucose influx of extracellular calcium. This may provide a new path for the study of can activate SOCC in rat BSMCs and enhance the diabetic cystopathy
