摘要
目的 观察大鼠杏仁核点燃癫痫后细胞凋亡情况。探讨其在点燃癫痫后不同脑区不同点燃时程时变化的意义。方洁 建立大鼠杏仁核点燃癫痫模型,应用流式细胞术(FCM)检测额叶、颞叶及海马神经细胞中DNA状态,同时应用TUNEL染色方法观察以上部位凋亡细胞变化。结果 大鼠点燃后1d既有凋亡细胞出现,点燃3d后凋亡细胞明显增多,并在1周时达高峰。点燃后21d及49d后,凋亡细胞有所减少,但仍高于正常水平,与对照组相比有显著意义。结论 凋亡现象贯穿于癫痫鼠病程的始终,与癫痫发作密切相关。
Objective To explore the significance of apoptosis of cells in different brain regions during dif-ferent phrases in epileptic rats kindled by electrical amygdala. Methods The model of epieptic rats was made by electrifed amygdala. DNA statement of neurons in frontal lobe, temporal lobe and hippocampus was opserved by flow cytometry. Meanwhile, apoptosis of neurons was observed by TUNEL assay. Results 1 day after kinding, the apoptosis appeared, increased obviously after 3 days, reached its peak,then attenuated after 21 days, but still above the normal level. Compared with control group, apoptosis in epileptic group increased evidently. Conclusion Apoptosis exists in the process of epilepsy. It may be closely related to epilepsy.
