纤维蛋白胶羊膜棒在去势雄兔干眼症的应用研究 被引量:2

摘要 目的研究纤维蛋白胶羊膜棒对去势雄兔干眼症的预防和治疗作用。方法选取雄性新西兰大白兔60只(60眼,均为右眼),行双侧睾丸切除术后随机分为A、B、C、D、E及F 6组(各组10眼)。A组手术后不进行任何处理观察6周;B组手术当日将纤维蛋白胶羊膜棒植入泪道5 s后取出观察6周;C组手术当日植入纤维蛋白胶羊膜棒6周;D组造模(手术2个月后形成干眼模型)后不进行任何处理观察6周;E组造模当日将纤维蛋白胶羊膜棒植入泪道5 s后取出观察6周;F组造模当日植入纤维蛋白胶羊膜棒6周。分别于植入前和植入后2、4及6周行Schirmer I试验(SIT)、角膜荧光素染色(FL)、角膜共聚焦显微镜扫描和泪道黏膜K16的Western blot检查。结果 C组在植入2、4及6周后SIT较植入之前好转,差异有统计学意义(P<0.05);C组与A、B组间的SIT在各时间比较,差异有统计学意义(P<0.05);在植入2周后,C组FL指标评分较植入之前无明显变化,差异无统计学意义(P>0.05);在植入4和6周后C组与A、B组FL指标评分较植入之前轻度好转,差异有统计学意义(P<0.05)。在植入2、4及6周后,F组SIT较植入前好转,差异有统计学意义(P<0.05),F组FL指标评分较植入之前好转,差异有统计学意义(P<0.05);植入后C、F组上皮基底细胞和炎症细胞均减少,差异有统计学意义(P<0.05)。免疫荧光染色及Western blot结果显示C、F组泪道黏膜K16阴性表达。结论纤维蛋白胶羊膜棒能够有效地预防和治疗去势雄兔干眼症,延缓干眼症的进展。 Objective To investigate the effect of fibrin glue amniotic rod on prevention and treatment ofdry eye in models of castrated rabbit. Methods A total of 60 castrated male rabbits (60 right eyes) wererandomly divided into: groupA (sham group), groupB (sham plus fibrin glue amniotic rod for 5 seconds),groupC (sham plus fibrin glue amniotic rod for 6 weeks), groupD (dry eye), groupE (dry eye plus fibringlue amniotic rod for 5 seconds) and groupF (dry eye plus fibrin glue amniotic rod for 6 weeks). Animalswere observed for 6 weeks after surgery. Schirmer I test (SIT), Corneal fluorescein (FL), Immunofluorescentstaining on cornea and Western blot analysis of K16 expression were performed at baseline, 2 weeks, 4 weeksand6 weeks after treatment. Results As for group C, SIT showed beneficial effect of fibrin glue amniotic rodsensitively on the rates at each recording time after treatment when compared with those in the baseline( 〈0.05). Group C had a significant improvement in terms of SIT in comparison with group A and B at varioustime points( 〈 0.05). Group C hada significant improvement in terms of FL in comparison with group A andB at the time point of 4 weeks and6 weeks after treatment( 〈 0.05). GroupF experienceda dramaticamelioration of SIT and FL at the time point of 2 weeks, 4 weeks, and6 weeks after treatment ( 〈 0.05).Epithelial basal cells and inflammatory cells were significantly decreased in group C andF ( 〈 0.05),Immunofluorescent staining and Western blot showed downregulated expression of K16 in lacrimal mucosa ingroup C and F when compared with other groups( 〈 0.05).
