穹隆海马伞切断大鼠海马PKC和ERK活性的变化

摘要 目的 探讨信号转导机制在阿尔茨海默病(Alzheimer’s disease,AD)发病过程中的作用。方法 应用脑立体定位仪制备穹隆海马伞切断AD大鼠模型,采用放射性同位素方法检测大鼠海马蛋白激酶C(protein ki-nase C,PKC)和细胞外调节蛋白激酶(extracellular regulated Protein kinase,ERK)的活性。结果 手术组手术侧海马PKC较正常对照组降低,手术组手术侧海马ERK较正常对照组升高。结论 在AD大鼠海马PKC和ERK的变化证实了信号转导机制在AD的形成中起到了一定的作用。 Objective To observe the function of signal transduction mechanism in Alzheimer'disease (AD). Methods Transecting of rat fimbtia-fornix to obtain the AD model and investigating activities of PKC and ERK of rat hippocampus by radioisotope. Results The activities of PKC of experimental group operating side hippocampus decreased compared to normal control group, and the activities of ERK in operating side hippocampus of expermen-tal group increased compared to normal control group. Conclusion Changes of PKC and ERK in AD rat hip-pocampus demonstrate signal transduction mechnism have effects on AD.
