竹沥对慢阻肺大鼠肺组织中AQP1、AQP4、AQP5、TNF-α和IL-6的影响 被引量:8

摘要 目的:观察中药竹沥对慢性阻塞性肺疾病(COPD)大鼠肺组织中水通道蛋白1(AQP1)、4(AQP4)、5(AQP5)及肿瘤坏死因子-α(TNF-α)、白介素6(IL-6)表达的影响,探讨中药竹沥对慢性阻塞性肺病患者化痰疗效的可能机制。方法:选用雄性Wistar大鼠30只,适应性饲养1周后随机分为空白组、模型组、干预组,每组10只,除空白组外,其他组均采用香烟熏结合弹性蛋白酶气管滴入法复制COPD大鼠模型,90d后,干预组进行鲜竹沥口服液灌胃10d,余两组进行生理盐水灌胃10d。干预结束后采用PCR法观察AQP1、AQP4、AQP5的表达,酶联免疫吸附法(ELISA)测定大鼠肺组织中TNF-α和IL-6的含量。结果:与空白组比较,模型组、干预组肺组织中AQP1、AQP4、AQP5浓度显著下降(P<0.01),TNF-α、IL-6浓度显著升高(P<0.01)。与模型组比较,干预组肺组织AQP1、AQP5浓度显著升高(P<0.01),AQP4、TNF-α和IL-6浓度显著下降(P<0.01),差异均有显著统计学意义。结论:竹沥对COPD的疗效可能与调节水通道蛋白1、5(AQP1)、(AQP5)的转运,抑制肿瘤坏死因子-α(TNF-α)、白介素6(IL-6)表达密切相关。 Objective: To observe the effect ofbamboo juice on lungtissue of rats with COPD in AQP1, AQP4, AQP5 , TNF- and IL-6, To explore the possible mechanism of COPD patients with phlegm effect of juice. Methods: 30 male Wistar rats, fed after 1 weeks were randomly divided into control group, model group and inter-vention group, 10 rats in each group, except the blank group, other groups were treated with cigarette smoking combined with elastaseintratracheal instillation of COPD rats model was established, 90 days later, the intervention group in xianzhuli oral liquid gavage for 10 days, the two other groups were saline for 10 days. After the intervention, the expression of AQP1, AQP4 and AQP5 were observed by PCR method. The levels of TNF- and IL-6 in lung tissue were determined by enzyme linked immunosorbent assay (ELISA). Results: Compared with the blank group? the concentrations of AQP1, AQP4 and AQP5 in the lung tissue of model group and intervention group were significantly decreased (P〈 0.01) , and the levels of TNF- and IL-6 were significantly higher (P〈0.01). Compared with the model group, the lung tissue AQP1 and AQP5 concentrations in the intervention group were signifi-cantly increased (P〈0.01) , AQP4, TNF- and IL-6 concentrations were significantly decreased (P〈0. 01) ,the difference was statistically significant. Conclusion: Bamboo juice have a curative effect of COPD may be related to the regulation of aquaporin 1 and aquaporin 5 (AQP1) , (AQP5) transporter, inhibition of tumor necrosis factor al-pha (TNF- alpha) , interleukin 6 (IL-6) expression is closely related to.
