三氧化二砷对A549细胞株增殖的抑制作用研究

Effect of arsenic trioxide on the inhibition of A549 proliferation

目的探讨三氧化二砷(As_2O_3)对人肺腺癌A549细胞增殖的抑制作用。方法设置对照组和实验组1~5号,分别用不同浓度As_2O_3(1、2、4、8、10μmol/L)干预体外培养的人肺腺癌A549细胞株,干预时间为24、48、72 h。采用四甲基偶氮唑盐(MTT)法检测各组细胞的吸光度(OD)值,统计分析不同干预浓度、不同干预时间As_2O_3对A549细胞的增殖的抑制作用。结果 As_2O_3干预24 h各组OD值比较,差异无统计学意义(P>0.05);而干预48、72 h各组OD值比较,差异均有统计学意义(P<0.05);As_2O_3干预48 h的细胞增殖的抑制作用强于72 h。结论 As_2O_3能抑制体外培养的人肺腺癌A549细胞增殖,该抑制作用呈浓度依赖性,且作用48 h抑制作用最强。

Objective To investigate effect of arsenic trioxide（As2O3） the inhibitionof A549 proliferation. Methods Cultrued A549 in vitro and set up one control group and five experimental groups and labeled the experimental groups from 1 to 5respectively with different concentration of As2O3（1,2,4,8,10 μmol/L）. The time of intervention was 24,48,72 h. Absorbance（OD）value was detected by methyl thiazolyl tetrazolium（MTT） assay. Results The difference was not statistically significant between different groups in 24 h（P〉0.05）,however,the difference between OD values in 48,72 h was statistically significant（P〈0.05）,and the inhibition effect of As2O3 in 48 h was better than 72 h. Conclusion This experiment shows that As2O3 can inhibit the proliferation of A549 in vitro,and the inhibition effect is concentration dependent,and the strongest effect is in 48 h.
