丹参酮IIA对成年大鼠睾丸扭转的保护作用和机制研究 被引量:4

摘要 目的:观察丹参酮IIA对成年大鼠睾丸扭转复位睾丸损伤的保护作用并初步探讨其机制。方法:对建立左侧睾丸扭转复位动物模型,将36只7周龄健康SD雄性大鼠随机分为3组:假手术组、生理盐水组、丹参酮IIA组,每组12只;分别于术后72小时、4周两批处死大鼠,每批每组6只,取左侧睾丸,行HE染色及组织损伤评分,Tunel染色,以及睾丸组织内Bcl-2蛋白检测(免疫印迹法),检测总活性氧(ROS),超氧化物歧化酶(SOD)、与丙二醛(MDA)含量,并测定大鼠血清睾酮水平。结果:与生理盐水组比较,丹参酮II给药组在术后72小时睾丸组织损伤明显减轻,细胞凋亡减少,Bcl-2表达升高;在术后4周发现给药组SOD、大鼠血清睾酮水平明显上升,而组织内ROS和MDA水平含量下降,差异均具有显著性(P<0.05)。结论:单次腹腔内注射丹参酮IIA对成年大鼠单侧睾丸扭转损伤具有保护作用,其机制在早期可能是通过抗凋亡作用,而在远期则可能发挥了抗氧化作用。 Objective: To observe the protective effect of tanshinone IIA on testicular torsion and return to testicular injury in adult rats and to explore its mechanism.Methods: 36 male SD rats aged 7 weeks were randomly divided into 3 groups: sham operation group,saline group and tanshinone IIA group,12 rats in each group.The rats were sacrificed at 72 hours and 4 weeks after operation.Six rats in each group and left testes were subjected to HE staining and histological injury score,Tunel staining,and Bcl-2 protein test(immunoblotting),reactive oxygen(ROS),superoxide dismutase(SOD) and malondialdehyde(MDA) were measured.Serum testosterone levels were measured as well.Results: Compared with the saline group,the testis tissue injury was significantly reduced in the tanshinone IIA group,the apoptotic cells were decreased and the expression of Bcl-2 was increased.The levels of SOD and the level of serum testosterone in rats was significantly higher than that in saline group(P 0.05).Conclusion: Single intraperitoneal injection of tanshinone II has protective effect on unilateral testicular torsion injury in adult rats,possibly through the anti-apoptotic effect in early stage,and may play an antioxidant role in the long term.
