杠柳毒苷对神经胶质瘤细胞增殖及凋亡的影响

摘要 目的观察杠柳毒苷对神经胶质瘤细胞增殖和凋亡的抑制作用。方法体外实验中,将不同质量浓度的杠柳毒苷作用于神经胶质瘤细胞,MTT法检测其对神经胶质瘤细胞增殖的抑制作用;DAPI荧光染色观察细胞核形态的变化;流式细胞术分析给药后神经胶质瘤细胞凋亡及细胞周期的变化情况;Western blot法检测给药后周期蛋白依赖激酶抑制剂P27的表达量。结果杠柳毒苷可以显著抑制神经胶质瘤细胞的生长增殖,且随着药物浓度的增加和作用时间的延长抑制作用也随之加强。结论不同浓度的杠柳毒苷可显著促进神经胶质瘤细胞的凋亡,在抑制细胞增殖的过程中,细胞周期蛋白依赖性激酶抑制剂P27的表达量显著升高。 Objective To observe the effect of periplocin on proliferation and apoptosis of glioma cells.Methods Glioma cells were treated by periplocin at different concentrations in vitro experiments.The inhibitory effects on the growth and proliferation of glioma cells were detected by MTT method.The morphological changes of cell nucleus were assayed by DAPI fluorescent staining.The apoptosis rate and the cell cycle of glioma cells induced by periplocin were examined by flow cytometry.The protein expression of cyclin-dependent kinase inhibitor P27 of glioma cells was detected by Western blot.Results The experiment results suggested that periplocin had significant inhibitory effect on the growth and proliferation of glioma cells,and the inhibitory rate increased with the increasing concentration and prolonging reaction time.Conclusion Periplocin at different concentrations can significantly promote the apoptosis of glioma cells and the protein expression of cyclin-dependent kinase inhibitor P27 increases markedly during the inhibition of proliferation.
