摘要
目的通过检测血液样本中ALX4基因启动子区甲基化状态,为大肠癌早期诊断寻找非侵入性的筛查方法。方法收集25例大肠癌患者和25例健康对照者的血液样本,使用亚硫酸氢钠提取血DNA,采用甲基化特异性聚合酶链反应(MSP)分析ALX4基因启动子区Cp G岛的甲基化情况。结果 25例大肠癌患者中有18例存在ALX4基因甲基化,25例健康对照组中4例存在甲基化。肿瘤组样本中启动子区甲基化检测的敏感性为72%,特异性为84%。肿瘤组与对照组之间ALX4基因启动子的甲基化状态差异有统计学意义(P<0.05)。结论 ALX4基因启动子区甲基化检测在大肠癌诊断中具有较高的敏感度和特异度,可能成为大肠癌早期筛查的血清学标志物。
Objective To develop a non-invasive screening method for early diagnosis of colorectal cancer by detecting methylation status of promoter region of ALX4 gene in blood samples.Methods 25 patients with colorectal cancer and 25 healthy control subjects blood samples were collected,which were used sodium bisulfite to extract the blood DNA,used methylation specific polymerase chain reaction (MSP) to analysis of ALX4 gene promoter CpG island methylation.Results Methylation of ALX4 gene was present in 18 of 25 patients with colorectal cancer,and methylation in 4 of the 25 healthy controls.The sensitivity of promoter methylation detection in tumor samples was 72% and specificity was 84%.The methylation status of ALX4 gene promoter was statistically significant between tumor group and control group (P < 0.05).Conclusion Detection of ALX4 promoter methylation has high sensitivity and specificity in the diagnosis of colorectal cancer,and it may be a serological marker for early screening of colorectal cancer.
