摘要
目的观察维生素D_3对耐氟康唑白念珠菌的体外抗菌活性。方法参考美国临床和实验室标准化研究所(CLSI)制定的M27-A3方案测定维生素D_3对白念珠菌的最低抑菌浓度(MIC)值;用棋盘法测定维生素D_3与氟康唑的联合抗真菌效果。结果维生素D_3单独对白念珠菌的MIC50和MIC90分别为1,2μg·mL^(-1)。维生素D_3与氟康唑联合应用时,对80%的耐氟康唑白念珠菌菌株(n=10)有协同效应,氟康唑的MIC值由≥64μg·mL^(-1)降为0.25μg·mL^(-1),未发现拮抗效应。结论维生素D_3在体外对白念珠菌具有良好的抗真菌活性,与氟康唑联合时能增强氟康唑对部分耐药白念珠菌的抗菌活性。
Objective To investigate the in vitro antifungal activity of vitamin D3 against fluconazole - resistant Candida albicans. Methods The minimal inhibitory concentrations (MIC) of vitamin D3 against Candida albican were assayed by Clinical and Laboratory Stan- dards Institute (CLSI) M27 -A3 microdilution method; the combined antifungal activity of vitamin D3 with fluconazole was evaluated by broth microdilution checkerboard technique system. Results The MIC50 and MIC90 of vitamin D3 alone against Candida albican were 1, 2 μg·mL^(-1)The combination of vitamin D3 and fluconazole had synergistic inhibitory activity against fluconazole - resistant Candida albican ( 80%, n = 10) and MIC of fluconazole decreased from 64 μg·mL^(-1) to 0.25μg·mL^(-1) and no antagonism was found. Conclusion Results indicate that vitamin D3 had powerful antifungal activity against Candida albican, and enhanced the activity of fluconazole against fluconazole -resistant Candida albican in vitro.
