摘要
目的:观察丹参酮ⅡA磺酸钠(sodium tanshinone Ⅱ A sulfonate,STS)对百草枯中毒大鼠血清丙二醛(malondialdehyde,MDA)、超氧化物歧化酶(superoxide Dismutase,SOD)水平的影响,探讨丹参酮ⅡA磺酸钠对百草枯中毒大鼠氧化应激损伤的保护作用。方法:90只Sprague-Dawley(SD)雄性大鼠随机分成对照组、百草枯组、STS组,每组30只。对照组腹腔注射生理盐水2 ml,百草枯组给予生理盐水稀释百草枯50 mg/kg至2 ml灌胃染毒,STS组在百草枯染毒后给予STS 25 mg/kg腹腔注射,连续注射3天。各组分别于制模后24小时、48小时、72小时分别处死大鼠各10只,心脏取血,酶联免疫法检测血清MDA、SOD浓度。结果:百草枯组大鼠各时间点血清MDA浓度较对照组显著升高,其中在48小时达高峰;STS组大鼠各时间点血清MDA浓度较百草枯组比较呈逐渐下降趋势(P<0.05)。百草枯组大鼠各时间点血清SOD浓度较对照组比较呈逐渐下降趋势;STS组大鼠各时间点血清SOD浓度较百草枯比较呈逐渐升高趋势(P<0.05)。结论:STS对百草枯中毒大鼠的氧化应激损伤有一定的保护作用。
Objective:To investigate the protective effects of tanshinone IIA sulfonate(STS)on Oxidative StressInjury rats with Paraquat Poisoning by detecting serum Malondialdehyde(MDA)and Superoxide Dismutase(SOD)inrats. Methods:Ninety healthy Sprague-Dawley(SD)male rats were randomly divided into control group,paraquatgroup,STS group equally. The control group was given 2 ml saline by intraperitoneal injection. The Paraquat group andSTS group were given saline diluted paraquat 2 ml(50 mg/kg)by Intragastric administration. The STS group was givenSTS(25 mg/kg)by intraperitoneal injection for three days after paraquat exposure before. Ten rats of each group weresacrificed after the model was completed at 24-hour,48-hour and 72-hours. Blood samples were taken from heartsdirectly. The levels of serum MDA and SOD were measured by Enzyme-Linked Immunosorbent assay(ELISA). Results:The serum concentration of MDA in the paraquat group was significantly higher than that in the control group. The peaktime point is 48-hour. The concentration of MDA in the STS group decreased gradually compared to the paraquat group atall the three times point(P〈0.05). The serum SOD concentration in the paraquat group was lower than that of thecontrol. The serum SOD concentration in the STS group was higher than that of the paraquat group(P〈0.05).Conclusion:STS has a protective effect on oxidative stress injury in paraquat poisoning rats.
