摘要
目的研究经典干细胞标志物CD34、PDGF-βR、VEGF-R_2在早孕6~8周蜕膜组织中的表达。方法提取6~8周早孕期人工流产术蜕膜组织(n=16),消化后选用CD34、PDGF-βR、VEGF-R_2单克隆抗体进行荧光染色,分别采用单染、双染及三染法,应用流式细胞仪技术分析上述经典干细胞标志物的表达及其相互关系。结果 6~8周早孕期蜕膜组织细胞中,(1)单染法证实CD34^+细胞为(0.81±1.07)%,PDGF-βR^+细胞为(9.94±7.36)%,VEGF-R_2^+细胞为(0.90±1.19)%;(2)双染法证实CD34^+/VEGFR2^-细胞(0.59±0.61)%,CD34^+/VEGF-R2^+细胞(0.16±0.10)%;(3)三染法证实CD34^+/VEGFR2^+/PDGF-βR^+细胞(0.09±0.06)%,CD34^+/VEGFR2^-/PDGF-βR^+细胞(0.15±0.13)%。结论早孕6~8周蜕膜组织中存在经典干细胞标志物CD34、PDGF-βR、VEGF-R_2阳性表达的细胞,存在少量的双阳性表达及三阳性表达的细胞。早孕蜕膜组织中存在干细胞参与其发育过程。
Objective To study the expression of CD34,PDGF-βR and VEGF-R2 in the human decidual tissue of 6-8 weeks gestation.Methods The decidua tissue collected following induced abortion during 6-8 weeks of gestation was digested to form a single cell suspension.The expression of CD34,PDGF-βR and VEGF-R2 monoclonal antibodies were analyzed by single,double,and triple staining method.Flow cytometry was used to analyze the expression of these markers and their correlation.Results (1) In the 6-8 weeks early pregnancy decidual tissue cells,the concentration of CD34 +cells was (0.81 ± 1.07)%,PDGF-[3R+ cells (9.94 ± 7.36)%,and VEGF-R2 + cells (0.90 ± 1.19)%.(2) The concentration of CD34 +/VEGFR2-cells was (0.59 ± 0.61) %,and CD34 + / VEGF-R2 + cells (0.16 ± 0.10) %.(3) The concentration of CD34 + / VEGF-R2 +/PDGF-[3R + cells was (0.09 ± 0.06) %,and CD34 + / VEGF-R2-/PDGF-βR + cells (0.15 ±0.13)%.Conclusion The expression of CD34,PDGF-βR and VEGF-R2 in the early pregnancy decidual tissue was found,and there were also small amount of double positive and triple positive expression of the cells.Existence of stem cells in decidual tissue in early pregnancy suggests that those cells may be involved in the development of the decidua.
