抗支糖浆对哮喘大鼠BALF中IL-4、IL-13与IFN-γ含量的影响 被引量:2

摘要 目的:观察抗支糖浆对哮喘大鼠支气管肺泡灌洗液(BALF)中IL-4、IL-13及IFN-γ含量的影响,探讨其治疗哮喘的作用机制。方法:48只雄性大鼠随机分为空白组、模型组、地塞米松组、抗支糖浆组四组,除空白组外,其余各组采用皮下注射10%OVA、2%氢氧化铝混合液致敏及2%OVA雾化激发的方法制作哮喘大鼠模型。各组分别用生理盐水、地塞米松及抗支糖浆进行灌胃,连续灌胃14天后取大鼠BALF,离心留取上清液。采用ELISA法测定BALF中IL-4、IL-13与IFN-γ的含量,并用HE染色法观察肺组织病理学改变。结果:与空白对照组比较,模型对照组BALF中IL-4、IL-13含量明显升高(P<0.05),IFN-γ含量明显降低(P<0.05);抗支糖浆组IL-4、IL-13含量跟模型组相比均明显降低(P<0.05),IFN-γ含量显著升高(P<0.05)。肺组织病理显示,抗支糖浆可以减少哮喘引起的炎细胞浸润、支气管粘膜充血、渗出物的生成。结论:抗支糖浆能有效治疗哮喘大鼠,其作用机制可能与调节哮喘大鼠IL-4、IL-13和IFN-γ水平有关。 Objective: To observe the effect of Kangzhi syrup on IL - 4, IL - 13 and IFN - γ in BALF of the asthma rats, and to investigate its mechanism for treating asthma. Methods : 48 male rats were randomly divided into the blank group, the model group, Dexamethasone group and Kangzhi syrup group. The mixture injection of 10% OVA and 2% A1 ( OH)3, and 2% OVA inhalation were used to made asthma models in the rats. After 14 days of corresponding interven- tion, the levels of IL -4, IL - 13 and IFN -γ in BALF were detected wtih ELISA method, and pathological changes of the lung tissues were observed with HE - staining method. Results : Compared to the blank group, the levels of IL - 4 and IL- 13 were increased significantly in BALF in the model group(P 〈0.05) ; and the level of IFN -γ was reduced significantly (P 〈0.05 ). The levels of IL-4 and IL- 13 were apparently decreased and IFN-γwas significantly in- creased in Kangzhi syrup group than those in the model group ( P 〈 0.05 ). The pathological observation showed that Kangzhi syrup can reduce the infiltration of inflammatory cells, the hyperemia and exudation of bronchial mucosa caused by asthma. Conclusion: Kangzhi syrup can treat asthma rats effectively, and the mechanism may be related to the regu- lation of IL -4, IL - 13, and IFN - γ levels.
