摘要
目的研究艾拉莫德(T-614)对Ⅱ型胶原诱导性关节炎(CIA)模型大鼠核因子(NF)-κB信号的影响。方法将40只大鼠随机分成空白组,模型组,甲氨蝶呤(MTX)对照组,T-614治疗组,每组10只。造模后,采用RT-PCR及Western印迹法检测T-614对CIA大鼠关节滑膜细胞NF-κB/P50、NF-κB/P65、IκBα基因及蛋白水平的影响。结果 T-614可下调CIA大鼠滑膜细胞NF-κB/P50、NF-κB/P65的表达,上调IκBα的表达,与模型组比较有统计学差异(P<0.01);T-614治疗组优于MTX对照组(P<0.01或P<0.05)。结论 T-614可通过降低CIA大鼠关节滑膜细胞NF-κB/P50、NF-κB/P65表达,升高IκBα表达以达到控制关节炎的作用。
Objective To study the effect of T-614[N-( 3-formylamino-4-oxo-6-phenoxy-4 H-chromen-7-yl) methanesulfonamide on nuclear factor( NF)-κB signal of articular synovial cells in collagen induced arthritis( CIA) model rats. Methods 40 rats were randomly divided into blank control,model control,methotrexate( MTX),T-614 treatment groups,each group with 10 rats.RT-PCR and Western blot were used to detect the expressions of NF-κB/P50,NF-κB/P65,IκBα in each group.Results T-614 down-regulated the expressions of NF-κB/P65 and NF-κB/P50,up-regulated the expression of IκBα in articular synovial cells of CIA rats( P〈0.01); the effects in T-614 treatment group were better than those of MTX control group( P〈0.01 or P〈0.05). Conclusions T-614 could lower the expressions of NF-κB/P50 and NF-κB/P65,upregulate the expression of IκBα in articular synovial cells of CIA rats,through these ways,it could control arthritis.
