白藜芦醇诱导HepG2细胞凋亡中线粒体差异蛋白鉴定 被引量:2

摘要 文中分离提取白藜芦醇处理的HepG2细胞线粒体蛋白质,以未处理HepG2细胞为对照,利用双向电泳技术分离差异蛋白,飞行时间质谱分析鉴定差异蛋白点.初步分析鉴定了4个显著性差异蛋白,其中驱动蛋白和着丝粒相关蛋白CENP-E在白藜芦醇处理的HepG2细胞中表达降低,证明白藜芦醇对细胞周期有调节作用;线粒体加工肽酶表达降低,线粒体核糖体蛋白L7/L12表达升高,说明白藜芦醇诱导HepG2细胞凋亡与其影响线粒体功能有关. The effects of resveratrol on HepG2 cells apoptosis were investigated. Mitochondrial proteins were extracted from control and resveratrol treated cells and separated by two-dimensional electrophoresis. The peptide mass finger-printing was analyzed by time-of flight mass spectrometer. Four significantly differentiated proteins were identified. Kinesin protein and centromere-associated protein E(CENP-E) were down-regulated in HepG2 cells treated with resveratrol,which testified function of resveratrol on cell cycle regulation. Down-regulation of mitochondrial processing peptidase and up-regulation of mitochondrial ribosomal protein L7/L12 in HepG2 cells treated with resveratrol suggested the relationship between apoptosis induced by resveratrol and mitochondrial dysfunction in HepG2 cells.
