新型月季叶枯病菌产毒条件及毒素化学成分分析

摘要 为深入揭示月季叶枯病菌(Pestalotiopsis clavispora)的致病机理,本试验研究了其最优产毒条件,测定了含毒滤液中蛋白质和可溶性糖含量,并借助气相色谱-质谱联用仪(GC-MS)对月季叶枯病菌所产毒素中的化学成分进行了分析。结果表明,不同产毒条件下病菌产毒能力存在明显差异,最佳产毒条件是:p H 7.0的查彼+液体培养基、培养温度25℃、昼夜交替条件、连续振荡(120 r/min)培养25 d。含毒滤液的蛋白质浓度为180μg/m L,可溶性糖含量为2 057.25μg/m L,主要化学成分为三氧硅烷、十甲基环五硅氧烷、十二甲基环六硅氧烷、1,4二甲氧基-6,7,8,9四氢环庚酮、戊二酸、氨基甲酸甲酯。 To deeply reveal the pathogenesis of Pestalotiopsis clavispora,the toxin production conditions were optimized,the contents of protein and soluble sugar in toxin filtrate were measured,and the toxin chemical compositions were analyzed by GC-MS. The results showed that the toxin production ability varied significantly under different conditions. The best toxin production conditions were Czapek liquid culture medium with p H value of 7. 0,25℃,day alternates with night,continuously shaking for 25 days with the rotational speed of 120 r/min. Moreover,the protein content of toxin was 180 μg/mL. The soluble sugar content of toxin was2 057. 25 μg/mL. The chemical compositions of toxin were tripropoxysilane,decamethyl cyclopentasiloxane,dodecamethyl cyclohexasiloxane,1,4 dimethoxy-6,7,8,9 tetraheptone,glutarate,methyl carbamate.
