角质细胞生长因子对人龈上皮细胞在钛金属表面附着的影响

摘要 目的:探讨角质细胞生长因子(KGF)对人龈上皮细胞在钛金属表面附着的影响效果。方法:取阻生牙拔除术患者的健康龈组织块,经过相关处理获取龈上皮细胞,将该细胞在纯钛片表面接种,在重组KGF浓度调配下处理,进而计数纯钛片表面附着的细胞数,测定纯钛片表面细胞的粘附力,比较接种后6 h、1 d、5 d、10 d、15 d时不同KGF浓度下纯钛片表面附着的龈上皮细胞数目,记录KGF浓度1 ng/m L与5 ng/m L在培养1 d、10 d时龈上皮细胞在纯钛片表面粘附力与平均荧光强度(AFI)。结果:接种后同一时间点,随着KGF浓度升高,龈上皮细胞数目明显增多(P<0.05),同一KGF浓度下,随着接种时间延长,龈上皮细胞数目明显增多(P<0.05);KGF浓度1 ng/m L与5 ng/m L时,培养1 d与10 d时龈上皮细胞在纯钛片表面粘附力与AFI比较,差异均有统计学意义(P<0.05),培养10 d时更高。结论:随着KGF浓度升高、接种与培养时间延长,龈上皮细胞数目明显增加,而且附着力也会随着KGF浓度升高而升高,在时间-浓度变化中,AFI也呈现随之增高趋势,可见KGF对人龈上皮细胞在钛金属表面附着影响较大,需加强重视。 Objective:To investigate the effect of keratinocyte growth factor(KGF) on adhesion of human gingival epithelial cells on titanium surface.Method:The gingival epithelial cells were obtained from the patients with impacted teeth,and the gingival epithelial cells were obtained by related treatment,they were inoculated on the surface of pure titanium sheets,processed under the concentration of recombinant KGF,the number of cells attached to the surface of pure titanium sheets was counted,and the adhesion force of the cells on the surface of pure titanium sheets was measured.The number of gingival epithelial cells adhered to pure titanium slices at different concentrations of KGF at 6 h,1 d,5 d,10 d and 15 d after inoculation were compared,the adhesive force and average fluorescence intensity(AFI) of gingival epithelial cells on pure titanium sheets at l ng/mL and 5 ng/mL of KGF concentration at 1 d and 10 d were recorded.Result:At the same time point after inoculation,the number of gingival epithelial cells increased significantly with the increase of KGF concentration(P〈0.05).At the same KGF concentration,the data of gingival epithelial cells increased significantly with the extension of inoculation time(P〈0.05).When the concentration of KGF was 1 ng/mL and 5 ng/mL,the adhesion force and AFI of gingival epithelial cells on pure titanium surface were compared at 1 d and 10 d,the differences were statistically significant(P〈0.05),and the higher at 10 d.Conclusion:With the increase of KGF concentration,inoculation and incubation time,the number of gingival epithelial cells increased significantly,and the adhesion also increased with the increase of KGF concentration,in the time-concentration change,AFI also showed a trend of increasing,it showed that KGF had a great influence on the attachment of human gingival epithelial cells on the surface of titanium,so we should pay more attention to it.
