摘要
目的探讨复方中药(苦黄汤)在大鼠原位肝移植急性排斥反应模型中抑制急性排斥反应(Acute Rejection,AR)的原因。方法采用Wistar-SD原位肝移植AR模型,将40只大鼠受体分组为生理盐水组(n=20)和苦黄汤组(n=20)。从肝移植术前第1d至术后1个月,灌胃给苦黄汤(1次/d),剂量为2ml/d。术后7d处死生理盐水组大鼠和苦黄汤组大鼠各10只,评估排斥反应指数(RAI值),且分离大鼠原代肝细胞进行苦黄汤对肝移植术后AR模型中的肝细胞的研究。结果与生理盐水组对比,苦黄汤组术后生存时间显著长(P=0.016)。苦黄汤组大鼠RAI值明显低于生理盐水组。与生理盐水组相比,术后7d分离的苦黄汤组大鼠肝细胞增殖能力显著增高。结论苦黄汤可维持或增强原位肝移植AR模型大鼠肝细胞的增殖能力,且可能与抑制AR功能间存在正相关。
Objective To explore the causes of traditional chinese medicine(Kuhuang decoction) suppress acute rejection(AR) in the acute rejection model after orthotopic liver transplantation(0 LT) in rats. Methods The model of OLT was adopted in40 rats(Wistar donors and recipients),divided into normal saline group(n=20) and Kuhung decoction(n =20). From 1 day to 1 month before the operation of liver transplantation,Kuhung decoction to gavage(1 time/day),the dose is 2 ml/day. Ten recipients in each group were sacrificed on postoperative day 7,evaluated the rejection response index(RAI). The study of Kuhuang decoction using on hepatocyte in the AR model after liver transplantation which was carried out by isolated rat protohepatocytes were in progress. Results Compared with the saline group,the survival time of the Kuhuang decoction was significantly longer(P=0.016).RAI of Kuhuang decoction group was significantly lower than the normal saline group. Compared with the saline group,the proliferation ability of liver cells was significantly increased in Kuhuang decoction at the seven days after operation. Conclusion Kuhuang decoction can maintain or enhance the proliferation ability of liver cells in AR model of orthotopic liver transplantation,there may be a positive correlation on inhibition of AR function.
