摘要
目的:探讨蚕茧提取物—丝胶对大鼠INS-1细胞Bcl-2蛋白表达的影响。方法:INS-1细胞分为三组,正常对照组、链脲佐菌素(STZ)损伤组和丝胶处理组。正常对照组细胞用不含药物的完全培养基培养;STZ损伤组细胞用完全培养基配制的STZ溶液(10mmol/L)培养24h;丝胶处理组细胞用完全培养基配制的同时含STZ(10mmol/L)和丝胶(300μg/ml)的溶液培养24h。采用Western Blotting法检测各组INS-1细胞Bcl-2蛋白的表达情况。结果:与正常对照组相比,STZ损伤组INS-1细胞Bcl-2蛋白的表达明显降低(P<0.05);与STZ损伤组比较,丝胶处理组细胞Bcl-2蛋白的表达明显升高(P<0.05)。结论:丝胶可能通过上调Bcl-2蛋白的表达抑制β细胞凋亡。
Objective:To investigate the effects of sericin on expression of Bcl-2 protein in rats’INS-1 cells.Methods:INS-1 cells were divided into normal control group,streptozotocin(STZ)-injuried group and sericin-treatment group.The INS-1cells in normal control group were cultured with complete medium without any medicine;the INS-1 cells in STZ-injuried group were cultured with complete medium containing 10mmol/L STZ;the INS-1 cells in sericin-treatment group were cultured with complete medium containing 10mmol/L STZ and 300μg/ml sericin.Western Blotting was used to detect the expression of Bcl-2 protein in INS-1 cells of each group.Results:Compared with normal control group,the Bcl-2 protein expression in INS-1cells of STZ-injuried group decreased obviously(P〈0.05).Compared with STZ-injuried group,the Bcl-2 protein expression in INS-1 cells of sericin-treatment group increased obviously(P〈0.05).Conclusions:Sericin may inhibit beta cell apoptosis by up regulating the expression of Bcl-2 protein.
