人参皂苷Rg1对局灶性脑缺血再灌注后大鼠海马微管相关蛋白表达的影响 被引量:3

The Effect of the Ginsenoside Rg1 on the Expression of the Microtubule-associated Protein in Rat Hippocampus Following Focal Cerebral Ischemia and Reperfusion

摘要 目的探讨人参皂苷Rg1是否可通过促进局灶性脑缺血再灌注大鼠海马微管相关蛋白(Doublecortin,DCX)而改善神经功能缺损体征。方法采用大脑中动脉栓塞法(MACO)制作局灶性脑缺血再灌注模型。雄性SD大鼠随机被分成假手术组(n=10)、模型组(n=10)和人参皂苷Rg1组(n=10)。模型组脑缺血再灌注后30min腹腔注射等体积的0.9%生理盐水,1次/d,人参皂苷Rg1组ip人参皂苷Rg1(50mg/kg,浓度为8g/L),1次/d。14d后对3组大鼠进行神经功能评分;采用免疫组织化学染色和Western-blot观察3组大鼠DCX在海马的表达。结果与模型组相比,人参皂苷Rg1组神经功能缺损程度评分下降(P<0.05);给予人参皂苷Rg1干预后,局灶性脑缺血再灌注大鼠海马SGZ(齿状回颗粒细胞下层)区DCX阳性细胞、海马区DCX蛋白水平显著增加(P<0.05)。结论人参皂苷Rg1可通过上调海马微管相关蛋白的表达而改善局灶脑缺血大鼠的神经功能缺损体征。 Objective To explore the effect of Ginsenoside Rgl on the neurological function deficit and expression of Doublecortin(DCX) in rat hippocampus following focal cerebral ischemia reperfusion. Methods Focal cerebral ischemia reperfusion model was made by the MACO method and all male rats were randomly divided into sham operation group(n = 10),model group(n= 10) and Ginsenoside Rgl group(n = 10). The model group was administered with the equal volume of normal saline once a day 30 minutes after cerebral ischemia reperfusion. The Ginsenoside Rgl group was treated with Ginsenoside Rg1 once a day. The does of Ginsenoside Rgl was 50 mg/kg and the concentration was 8 g/L.Fourteen days later, the neurological function deficit score was assessed and the expression of DCX in rat hippocampus of 3 groups was observed via immunohistochemistry and Western-blot.Results In comparison with the model group, the neurological function deficit score of the Ginsenoside Rgl group decreased(P0. 05). After the intervention of Ginsenoside Rg1,the DCX positive cells in the hippocampal SGZ zone and the DCX protein level expressed in rat hippocampus significantly increased(P0. 05).Conclusion Ginsenoside Rgl could improve the neurological function deficit signs of rats with focal cerebral ischemia through upregulating the DCX expression in hippocampus.
