摘要
以黄花败酱皂苷提取物为原料,吸附率与洗脱率为衡量指标,考察5种大孔树脂对黄花败酱提取物中皂苷类化合物的静态吸附与洗脱性能,结合静态吸附动力学研究,确定选择AB-8型大孔树脂纯化黄花败酱提取物中皂苷类化合物。利用动态吸附-洗脱试验,得出最佳纯化工艺条件为:配置0.30 mg/m L的黄花败酱提取液,以2 BV/h流速,上样至径高比为1∶10的AB-8型树脂中吸附,随后采用5 BV的70%乙醇溶液,以2 BV/h流速洗脱。经高效液相色谱法定量分析表明,黄花败酱提取物中总皂苷含量由纯化前350.6 mg/g提高至纯化后846.9 mg/g。
In order to find the most appropriate macroporous resin to purify total saponins from Patrinia Scabiosaefolia Fisch extract. The static adsorption and desorption performance of five kinds of macroporous resins and combined with the kinetics of static adsorption for total saponins from Patrinia Scabiosaefolia Fisch extract were investigated by different adsorption and desorption ratios. AB-8 resin was found to be suitable for the purification of total saponins from Patrinia Scabiosaefolia Fisch by using dynamic adsorption and desorption experiments, the optimum purification conditions were obtained as follows: 0.3 mg/mL Patrinia Scabiosaefolia Fisch extract was loaded on to the resin column which the ratio of diameter-high was 1 : 10 at flow rate of 2 BV/h and eluted with 5 BV of 70 % ethanol at flow rate of 2 BV/ h. Under these conditions, the content of total saponins of Patrinia Scabiosaefolia Fisch extract was increased from 350.6 mg/g to 846.9 mg/g by quantitative analysis of high performance liquid chromatography.
