聚酰胺树脂纯化玫瑰花总黄酮工艺研究 被引量:6

摘要 优化聚酰胺树脂纯化玫瑰花总黄酮的工艺。采用紫外分光光度法,以芦丁含量为指标,测定玫瑰花提取物中总黄酮的含量。用单因素考察和正交试验相结合的方法筛选出聚酰胺树脂纯化玫瑰花总黄酮的最优工艺参数,对玫瑰花总黄酮进行富集纯化。聚酰胺树脂富集玫瑰花总黄酮的最优工艺条件为:采用质量浓度为4 mg/m L、pH5的上样液,径高比为1∶7,在室温下上样,吸附3 h以上,达到吸附饱和;解吸时先用15%乙醇洗去杂质,再用pH值为5~6的80%乙醇8 BV洗脱。由80%乙醇洗脱液得到的粉末中总黄酮含量达到23.4%,是纯化前的5.7倍左右。 To optimize the process of purifying total flavonoids in rose with polyamide resin. The content of total flavonoids in roses extract was determined by UV spectrophotometry with rutin content as an index. The total flavonoids in rose were enriched and purified by the optimal process parameters that screened by single factor and orthogonal test. The optimum conditions were as follows: the concentration and pH of sample solution were 4 mg/mL and 5; the ratio of diameter to height was 1 ∶ 7; the temperature of loading sample was room temperature; reaching adsorption saturation within over 3 h;scouring off impurity with 15 % ethanol; washing with 80 % ethanol (pH 5-6,8BV). The content of total flavonoids in the dried powder was rised to 23.4 %,which was about 5.7 times as much as before purification.
