聚维酮碘溶液对钛板表面金黄色葡萄球菌细菌生物膜的体外抗菌活性

In vitro bactericidal effect of povidone iodine solution on bacterial biofilm of staphylococcus aureus

摘要 目的观察两种不同浓度的聚维酮碘溶液对金黄色葡萄球菌(SA)细菌生物膜(BBF)的体外抗菌活性。方法在45枚钛板表面建立SA的BBF后,将其随机分成3组,每组15枚钛板。其中两组随机选取10枚钛板分别置于0.25%、0.5%浓度的聚维酮碘溶液中作用5 min,另外一组作为空白对照组;各组中的其余5枚钛板经超声裂解后做细菌培养,观察是否有细菌生长。3组活菌率比较以SPSS 17.0软件采用单因素方差分析,组内不同层的活菌率比较采用t检验。结果对照组培养24 h后成熟BBF中层活菌率为(67.4±1.1)%,底层活菌率为(39.4±2.2)%。中层和底层的BBF活菌率具有明显的统计学意义(t=35.81,P<0.01)。0.5%、0.25%聚维酮碘溶液作用5 min后的BBF的中层、底层活菌率分别为(66.7±1.4)%,(38.2±3.2)%、(66.5±1.0)%,(39.1±2.4)%;0.5%聚维酮碘溶液作用5 min后的BBF的中层、底层活菌率与0.25%聚维酮碘溶液作用5 min后的BBF的中层、底层活菌率和空白对照组相比差异都无统计学意义(P>0.05)。对照组、0.25%和0.5%聚维酮碘组钛板经超声裂解后的血平板细菌培养均可见SA菌落形成。结论 0.25%、0.5%浓度的聚维酮碘溶液对钛板表面SA的BBF作用5 min无明显抗菌活性。 Objective To explore in vitro bactericidal effect of povidone iodine solution of different concentrations on bacterial bio-films of staphylococcus aureus. Methods Staphylococcus aureus bio-film models were established in the 45 pieces of titanium plate surface and then all the titanium plates were divided randomly into three groups( 15 for each group). Ten titanium plates of the two groups were immersed in 0. 5%,0. 25% of povidone iodine solution respectively for 5 min,the third group was the blank control group. Five other pieces of titanium plates of the three groups after sonication treatment were used for bacterial cultivation to observe the bacterial growth. The living bacterial rates were analyzed by t test or chi-squaretest by SPSS 17. 0. Results After 24 h of incubation in the control group,the living bacteria rate at the middle layer of bio-film of the control group was( 67. 4 ± 1. 1) %,at the bottom the living bacteria rate was( 39. 4 ± 2. 2) %. The living bacteria ratio difference of bacterial bio-film between the middle layer and bottom layer of the control group was statistically significant( t = 35. 81,P〈0. 01).After being immersed in 0. 5% povidone iodine solution,the living bacteria rate of middle bio-film was( 66. 7 ± 1. 4) % and that of the bottom bio-film was( 38. 2 ± 3. 2) %. After being immersed in 0. 25%povidone iodine solution,the living bacteria rate of middle bio-film was( 66. 5 ± 1. 0) % and that of the bottom was( 39. 1 ± 2. 4) %. Comparing the groups of 0. 25% and 0. 5% povidone iodine solutions and the control group,the living bacteria rates of middle biofilm and the bottom showed no statistical significance respectively( all P〈0. 05). Bcterial cultivation of the control group,0. 25% and 0. 5% povidone iodine solution groups showed staphylococcus aureus colony. Conclusion The iodine solutions of 0. 25%,0. 5%concentrations which immerse the titanium plate surface with bacterial bio-film of staphylococcus aureus for5 min present no obvious antibacterial activity.
