碱液浸提龙胆多糖及化学组成分析

摘要 以龙胆为原料,研究龙胆多糖的浸提参数并进行多糖组成分析。以料液比、浸提温度、碱液浓度、浸提时间为因素,以多糖得率为指标,进行单因素试验和正交试验,研究碱液浸提龙胆多糖的最佳工艺参数。结果表明,各因素对龙胆多糖得率影响的主次顺序为浸提温度>碱液浓度>浸提时间>料液比,最佳参数为料液比1∶25,浸提温度80℃,浸提时间3 h,碱液浓度0.6 mo L/L,龙胆多糖得率为14.3%。通过龙胆多糖的化学组成分析可知总糖含量为85.2%,蛋白质含量为11.6%。 Gentian of Gentianaceae was the dried root and rhizome of the Gentiana manshurica, Gentian, Gentiana Triflora pall or Gentiana rigescens, which was commonly used in traditional Chinese medicine because of its bitter, cold, clearing damp heat in the liver and gallbladder,purgative coke stagnated fire. In this experiment, the water extracts of Gentiana residue as the raw material,the extraction parameters and chemical composition were investigated Firstly,the solid-liquid ratio,extraction temperature,alkali concentration,leaching extraction time were studied as the factors of single factor and orthogonal test of alkali leaching optimum parameters for gentian polysaccharide. The results showed that the various factors of Gentian polysaccharide yield effect of primary and secondary order was temperature > alkali concentration > time > solid-liquid ratio. The optimal parameters were solid-liquid ratio 1 ∶25,leaching temperature 80 ℃, extraction time 3 h, alkali concentration 0.6 mo L/L. The yield of gentian polysaccharide was 14.3%. The chemical compositions of polysaccharide were 85.2% total sugar and 11.6% protein.
