凝血与补体级联反应信号通路对艾滋病湿热内蕴证发病的影响 被引量:9

摘要 目的明确艾滋病湿热内蕴证蛋白水平的表达差异,探索凝血与补体级联反应信号通路在中医证候实质中所主导的作用。方法 2014年8月根据整群随机抽样原则,纳入艾滋病患者48例(艾滋病肺脾气虚证25例为艾滋病肺脾气虚证组,艾滋病湿热内蕴证23例为艾滋病湿热内蕴证组),并以同地区人口资料相似的非HIV感染者16例为对照组,采集所有受试者血清进行i TRAQ技术研究,将差异蛋白进行通路注释。结果艾滋病湿热内蕴证组与艾滋病肺脾气虚证组相比,α1-抗胰蛋白酶、补体C4结合蛋白(C4bp)表达下调;艾滋病湿热内蕴证组与对照组相比,α1-抗胰蛋白酶、补体C4bp、α-2巨球蛋白、人结合丝氨酸凝集素蛋白酶2表达上调,补体C1r subcomponent、补体因子B及载脂蛋白B variant表达下调。这些差异蛋白均存在于凝血与补体级联反应信号通路。结论艾滋病湿热内蕴证的发病机制可能是通过凝血与补体级联反应信号通路实现的,该信号通路上的差异蛋白点可作为艾滋病临床治疗的潜在用药靶点。 Objective To make clear the expression differences of the protein levels in the syndrome of interior dampness - heat of AIDS,and explore the role of signal path of coagulaton and complement cascades reaction in the nature of TCM syndrome. Methods By random cluster sampling principle,48 AIDS patients(25 with syndrome of Qi deficiency of lung and spleen of AIDS was Qi deficiency of lung and spleen of AIDS group,23 with syndrome of interior dampness - heat of AIDS was interior dampness - heat of AIDS group)were enrolled in October 2014,and non HIV infected individuals with similar demographic data in the same area were taken as the control group. The serum samples of all the subjects were studied with iTRAQ techniques,and the differential proteins were annotated by signal pathway. Results Compared with Qi deficiency of lung and spleen of AIDS group,the expression of α1 - antitrypsin and complement C4bp was down - regulated in interior dampness - heat of AIDS group. Compared with the control group,the expression of α1 - antitrypsin,complement C4bp,α - 2 macroglobulin and human - binding serine lectin protease 2 was up - regulated and the expression of C1r subcomponent complement,complement factor B,apolipoprotein B variant was down - regulated in interior dampness - heat of AIDS group. These proteins activated in coagulation and complement cascades reaction signal path. Conclusion The pathogenesis of AIDS patients suffering from syndrome of dampness - heat may be achieved through coagulation and complement cascades reaction signal path,and the differential proteins in this signal path can be used as potential drug targets for clinical treatment of AIDS.
