车前子黄酮的响应面法优化微波提取及其对鸡抗氧化能力的影响 被引量:13

Optimization of microwave-assisted extraction of plantain seed flavonoids by response surface methodology and its effects on antioxidant activities in chickens response to oxidative stress

摘要 本试验旨在单因素试验的基础上,考察乙醇体积分数、料液比、微波处理时间对车前子黄酮提取率的影响,利用响应面法确定微波提取车前子黄酮的最佳工艺,并研究微波提取的车前子黄酮对环磷酰胺(cyclophosphamide,CY)诱导鸡氧化应激的影响。选用100只1日龄白羽种蛋鸡公雏,随机分为5组(每组20只),即空白组、CY组(环磷酰胺+生理盐水)组、高剂量组(环磷酰胺+60 mg/mL车前子黄酮)、中剂量组(环磷酰胺+40 mg/mL车前子黄酮)和低剂量组(环磷酰胺+20 mg/mL车前子黄酮),比较不同水平车前子黄酮对氧化应激鸡抗氧化能力的影响。结果显示,车前子黄酮最佳提取条件是乙醇体积分数为64.80%、料液比为1∶21.58、微波处理时间为7.08 min,该条件下黄酮的提取率为11.40 mg/g。动物试验结果表明,车前子黄酮可显著提高氧化应激鸡血清的总抗氧化能力,提高超氧化物歧化酶、谷胱甘肽过氧化物酶和过氧化氢酶的活性,并可以降低血清中丙二醛的浓度。结果表明,采用响应面法对车前子黄酮提取条件进行优化合理可行,车前子黄酮可在一定程度上提高鸡机体的抗氧化能力。 Based on single-factor experiments(ethanol concentration,solid-to-liquid ratio and microwave treatment time),microwave-assisted extraction of flavonoids from plantain seed(PSF) was optimized using the response surface methodology.The effects of PSF on antioxidant activities were preliminarily evaluated in immunosuppressed chickens.Total 100 male chicks were randomly divided into 5 groups.Five groups were blank group,cyclophosphamide(CY) group,high-dose group(CY+60 mg/mL PSF),middle-dose group(CY+40 mg/mL PSF),and low-dose group(CY+20 mg/mL PSF).Different levels of PSF were compared on the effects of antioxidant activities in CY immunosuppressed chickens.The results indicated that the optimal extraction conditions with a yield of 11.40 mg/g for extraction of flavonoids(PSF) were ethanol concentration 64.80%,liquid-to-solid ratio 1 ∶ 21.58,microwave treatment time 7.08 min.In vivo antioxidant activity assay further showed that PSF increased the total antioxidant capacity(T-AOC) and the activities of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),catalase(CAT),and decreased the concentration of malonaldehyde(MDA) in immunosuppressed chickens induced by cyclophosphamide.These results suggested that PSF could have potential antioxidant applications as medicine.
