摘要
采用高效液相色谱法测定秸秆发酵液中L-乳酸与D-乳酸单体含量。通过正交试验优化色谱使用条件,以0.5 g/L硫酸铜溶液为流动相,柱温40℃,流速为0.7 mL/min。在此条件下,L-乳酸与D-乳酸分离度好,出峰时间短,峰面积大。乳酸的质量浓度0.10~100 mg/L范围内,与峰面积呈线性关系。该方法检出限(3N/S)为0.05 mg/L。样品中L-乳酸与D-乳酸的加标平均回收率分别为104.4%和100.5%,测定值的相对标准偏差(n=6)分别为1.3%和1.8%。
The contents of L-lactic acid and D-lactic acid monomer in straw fermented broth were determined by HPLC.The chromatographic conditions were optimized by orthogonal test.The 0.5 g/L copper sulfate solution was used as the mobile phase.The column temperature was 40℃ and the flow rate was 0.7 mL/min.Under these conditions,the separation degree of L-lactic acid and D-lactic acid is good,the peak time is short and the peak area is high.The mass concentration of lactic acid in the 0.10-100 mg/L range is linearly related to the peak area.The detection limit(3 N/S) of this method is 0.05 mg/L.The average recoveries of L-lactic acid and D-lactic acid in the sample were 104.4% and 100.5%,respectively.The relative standard deviations(n=6) of the measured values were 1.3% and 1.8%,respectively.
