摘要
目的探讨阿霉素联合热疗、程序死亡受体配体1抗体对胶质瘤细胞的增殖抑制作用,以及阿霉素联合热疗对U251胶质瘤细胞表面程序死亡受体配体1表达的影响。方法采用CCK-8法确定阿霉素的半抑制浓度。建立U251与T细胞共培养体系,采用CCK-8法检测不同处理组(阿霉素组,热疗组,程序死亡受体配体1抗体组,阿霉素联合热疗组,阿霉素联合程序死亡受体配体1抗体组,热疗联合程序死亡受体配体1抗体组,阿霉素联合热疗、程序死亡受体配体1抗体组)的细胞增殖抑制率。采用流式细胞术检测阿霉素联合热疗对U251胶质瘤细胞表面程序死亡受体配体1表达的影响。结果阿霉素联合热疗、程序死亡受体配体1抗体相比单一治疗方法抑制细胞增殖能力更强;阿霉素联合热疗可以上调胶质瘤细胞表面程序死亡受体配体1的表达。结论阿霉素联合热疗、程序死亡受体配体1抗体是一种很有前景的联合治疗胶质瘤方式,可为未来进一步探索胶质瘤治疗新策略提供实验依据。
Objective To observe the effect of adriamycin combined with thermotherapy and anti-programmed cell death ligand-1(PD-L1)antibodies on cell proliferation in U251 glioma cells, and to explore the expression of PD-L1 of U251 glioma cells after being treated with adriamycin and/or thermotherapy.Methods IC50 of adriamycin for U251 cells was determined by CCK-8 assay.The U251 cells were divided into several groups according to different treatments,then the viability of cells were measured by CCK-8 assay.The expression of PD-L1 on the surface of U251 cells was determined by flow cytometry after treated with adriamycin and/or thermotherapy. Results Adriamycin combined with thermotherapy and anti-PD-L1 antibodies could inhibit glioma cells growth obviously compared with other groups. Adriamycin combined with thermotherapy could up-regulate the expression of PD-L1.Conclusion Adriamycin combined with thermotherapy and anti-PD-L1 antibodies is a promising treatment regime for glioma and our research provides theoretical basis for the combination of adriamycin,thermotherapy and immunotherapy in glioma treatment.
