摘要
目的探讨增加miR-502表达对大肠癌细胞功能的影响。方法将大肠癌细胞HCT116分为实验组、对照组。实验组细胞转染miR-502 mimics,对照组未转染miR-502mimics,采用MTT方法检测miR-502对HCT116增殖的影响;平板克隆形成实验观察转染miR-502后HCT116细胞集落形成情况,Transwell实验检测miR-502对大肠癌细胞迁移和侵袭的影响。结果 MTT实验结果显示,与对照组相比,转染miR-502 mimics的HCT116第1~5天的吸光度值减低,差异有统计学意义(P<0.001)。在HCT116细胞的克隆实验中,实验组和对照组间细胞集落数差异有统计学意义(P<0.001),实验组细胞集落形成数量较对照组少,集落体积明显小于对照组。Transwell实验结果显示,与对照组相比,转染miR-502 mimics后,HCT116细胞的迁移及侵袭能力明显下降(P<0.001)。结论 miR-502表达上调可抑制HCT116生长、迁移及侵袭能力,miR-502有潜在抑癌作用。
Objective To explore the effect of miR-502 expression on colorectal cancer cells. Methods The colo-rectal cancer cells HCTll6 were divided into experimental group and control group, miR-502 mimics were trans- fected to HCTll6 in the experimental group but not transfected to HCTll6 in the control group. The effect of miR- 502 on the proliferation of HCT116 cells was detected by MT-F method. The colony formation of HCT116 cells was observed by plate-cloning assay after transfection of miR-502 mimics. The effect of miR-502 on the migration and invasion of colorectal cancer cells was evaluated by Transwell assay. Results The optical density value of HCT116 MTI" assay was significantly lower in the first day to the fifth day than that in the control group. The difference was statistically significant (P 〈 O. 001 ). In the cloning experiment of HCT116 cells, there were significant differences in the number of colonies between the experimental group and the control group (P 〈 0. 001 ). The number of colo- ny formation in the experimental group (miR-502mimics group transfected) was less than that in the control group and the colony volume was significantly smaller than that in the control group. The results of Transwell showed that compared with the control group, the migration and invasion ability of HCT116 cells were significantly decreased af- ter miR-502 mimics transfection(P 〈 0. 001 ). Conclusion The up-regulation of miR-502 can inhibit the growth, migration and invasion of HCTll6. MiR-502 has potential anti-cancer effect.
