摘要
目的通过建立肺炎支原体(MP)感染BALB/C小鼠模型,探讨克拉霉素对MP感染小鼠模型血清中和支气管肺泡灌洗液中的干扰素-γ(IFN-γ)、白细胞介素-6(IL-6)和白细胞介素-17(IL-17)表达水平的影响。方法 60只无特殊病原体(SPF)级6周龄BALB/C小鼠(17~20 g)按随机数字表法分为正常组、MP感染组和克拉霉素治疗组,每组20只。感染组及克拉霉素治疗组小鼠鼻内接种50μl含1×106 cfu/ml的MP菌液,建立MP感染模型,正常组接种等量0.9%氯化钠注射液。成功建立MP感染模型后,治疗组用克拉霉素治疗5 d。酶联免疫吸附测定法(ELISA)和反转录实时荧光定量-聚合酶链反应法(RT-PCR)检测3组小鼠血清及支气管肺泡灌洗液中IFN-γ、IL-6、IL-17 m RNA及蛋白的表达。结果与正常组相比,MP感染组小鼠血清及支气管肺泡灌洗液中IFN-γ、IL-6、IL-17 m RNA及蛋白的水平显著升高(P<0.05)。克拉霉素治疗后,小鼠血清及支气管肺泡灌洗液中IFN-γ、IL-6、IL-17 m RNA及蛋白的表达水平显著低于MP感染组(P<0.05)。结论克拉霉素可显著降低MP感染小鼠血清及支气管肺泡灌洗液中IFN-γ、IL-6、IL-17的水平。
Objective To establish the model of BALB/C mice infected with mycoplasma pneumonia (MP), and to investigate the effect of Clarithromycin on the expression levels of interferon-γ(IFN-γ), interleukin-6 (IL-6) and IL-17) in serum and bronchoalveolar lavage fluid of mice infected with MP. Methods Sixty 6-week-old specific pathogen free (SPF) BALB/C mice (17-20 g) were randomly divided into the normal group, MP infection group and clarithromycin treatment group (n=20 each). The mice in the infection group and clarithromycin treatment group re-ceived 50 μl 1&#215;106 cfu/ml MP solution by intranasal inoculation, and the MP infection model was established. The normal group was given the same equivalent normal saline. After the successful establishment of the MP infection model, the treatment group was treated with clarithromycin for 5 d. The mRNA and protein expression of IFN-γ, IL-6 and IL-17 in the serum and bronchoalveolar lavage fluid were determined by enzyme-linked immunosorbent assay (ELISA) and real-time quantitative PCR (RT-PCR). Results Compared with the normal group, the mRNA and protein expression levels of IFN-γ, IL-6, IL-17 in the serum and bronchoalveolar lavage fluid of MP-infected mice significant-ly increased (P〈0.05). The mRNA and protein expression levels of IFN-γ, IL-6 and IL-17 in the serum and bron-choalveolar lavage fluid of mice treated with clarithromycin were significantly lower than those in the MP infection group (P〈0.05). Conclusion Clarithromycin may significantly reduce the levels of IFN-γ, IL-6 and IL-17 in serum and bronchoalveolar lavage fluid of mice infected with MP.
