HER-2、GH/GHR在胃癌患者血清与肿瘤组织中的表达及其在胃癌发生发展过程中的作用

Expression and Effect of HER-2 and GH/GHR in Serum and Tumor Tissue of Patient with Gastric Cancer

目的探讨人表皮生长因子受体(HER-2)、生长激素(GH)/生长激素受体(GHR)在胃癌患者血清与肿瘤组织中的表达及其在胃癌发生发展过程中的作用。方法收集62例胃癌患者的血标本、术后癌组织及癌旁组织,同时收集同期50例健康者的血标本,采用酶联免疫吸附法(ELISA)检测血清中HER-2、GH的表达,采用免疫组化(IHC)法检测癌组织及癌旁组织中HER-2、GHR的表达。结果 HER-2、GH在胃癌患者血清中的阳性表达率均明显高于健康者(P<0.05);HER-2在胃癌患者癌组织中的阳性表达率明显高于癌旁组织,而GHR阳性表达率明显低于癌旁组织(P<0.05)。HER-2与GH在胃癌患者血清中的表达呈显著正相关(P<0.05),在胃癌患者癌组织中的表达亦呈显著正相关(P<0.05)。GH在胃癌患者血清中的表达阳性与淋巴结转移有一定的关系(P<0.05);HER-2在胃癌患者癌组织中的阳性表达与淋巴结转移、TNM分期有一定的关系(P<0.05),而GHR与分化程度有一定的关系(P<0.05)。结论HER-2与GH/GHR在胃癌患者血清与肿瘤组织中的表达存在紧密联系,可能在胃癌的发生、发展及转移过程中起着协同作用。

Objective To investigate the expression of HER-2 and GH/GHR in serum and tumor tissue of patient with gastric cancer and their effect on the development of gastric cancer. Methods The blood specimens, postoperahve carcinoma tis- sues and adjacent tissues of 62 cases patients with gastric cancer were collected, and blood specimens of 50 cases healthy people in the corresponding period were collected. HER-2 and GH in serum were detected by ELISA. and HER-2 and GHR in carcinoma tissues and adjacent tissues were detected by IHC. Results The positive expression rate of HER-2 and GH in serum of patients with gastric cancer was significantly higher than healthy people（ P 〈 0.05 ）. The positive expression rate of HER-2 m carcinoma tissues was significantly higher than adjacent tissues （P 〈 0, 05 ） , while GHR in carcinoma tissues was significantly lower than ad- jacent tissues（ P 〈 0.05 ）. The express{on of HER-2and GH in serum of patients with gastric cancer had positive significant corre- lation（ P 〈 0.05 ）, and in carcinoma tissues also had positive significant correlation（ P 〈 0.05 ）. The relationship, positive expres- sion of GH in serum of gastric cancer had a certain relationship with lymph node metastasis （ P 〈 0.05 ） .The positive expression of HER-2 in carcinoma tissues had a certain relationship with lymphatic metastasis and TNM staging （ P 〈 0.05 ） ,, and GHR had a certain relationship with differentiation degree （ P 〈 0.05 ）. Conclusion The expression of HER-2 and GH/GHR in serum and tumor tissue of patient with gastric cancer are closely correlated, it maybe play a synergistic effect on the occurrence, development, and metastasis of gastric cancer.
