摘要
研究根系的材料力学特性时,根系一般处于离体状态,对于柠条、沙柳、沙棘、白沙蒿4种植物根系离体后的存活时间和存活状态的研究及保存方法的探讨目前还未见报道,所以确定植物根系材料力学特性研究的适宜保存时间及保存方法亟待解决。本研究采用TTC-脱氢酶还原法测定1~4 mm径级范围内的上述4种植物根系在4℃低温培养箱中保存0天、6天、13天、20天的根系活性值。结果表明:4℃低温培养箱更适合较长时间保存植物根系活性;4种植物的根系活性减小率均随保存时间的增加而快速增长,当保存6天时,4种植物的根系活性减小率达到10%~20%,当保存20天时,4种植物的根系活性减小率达到70%~90%;保存时间相同时,在1~4 mm根径范围内,4种植物基本均随根径的增加根系活性减小率变小,即细根的根系活性更易损失。
When studying the mechanical properties of roots,the roots are generally in the isolated state.At present,there are no reports on the survival time,survival status and preservation method of roots of 4 plant species of Caragana korshinskii Kom,Salix psammophila Schneid,Hippophae rhamnoides Linn,Artemisia sphaerocephala Krasch in vitro.Therefore,it is urgent to determine the suitable storage time and the preservation method for the study on the mechanical properties of plant roots.In this research,TTC-dehydrogenase reduction method was used to determine the root activity of the 4 plant species in the 1-4 mm diameter range in low temperature incubator of 4℃ for 0 days,6 days,13 days and 20 days.The results showed that:low temperature incubator of 4℃ was more suitable for the preservation of plant root activity for a long time,the decrease rate of root activity of the 4 plant species was rapidly increased with increasing storage time;when roots stored for 6 days and 20 days,the decrease rate of root activity of the 4 plant species achieved 10%-20%,and 70%-90%,respectively;when the storage time was the same,the decrease rate of root activity of the 4 plant species was reduced with the increase of root diameter in the range of 1-4 mm,namely,the root activity of fine roots was more likely to be lost.
