摘要
目的探讨S100B在急性非特异性脊髓炎患者脑脊液中的表达变化及S100B对急性非特异性脊髓炎(ANM)患者病情评估的意义。方法选取2015年8月至2016年8月武装警察部队后勤学院附属医院脑科中心收治的8例ANM患者为研究对象,选取脑脊液检测未发现异常的正常脑脊液作为对照组,采用蛋白质组学技术检测脑脊液中S100B的表达变化,并通过酶联免疫吸附测定方法验证对照组及ANM患者治疗前后脑脊液中S100B的表达情况。结果 ANM患者急性期磁共振成像检查可发现多发性高密度影,经治疗后患者症状改善,弥散性高密度病灶消失。蛋白质组学技术及酶联免疫吸附测定方法均可以检测到脑脊液中S100B的存在,其中蛋白质组学为S1∶S2=0.33;S1∶N=9.60;放射免疫测定S100B:与对照组比较,ANM患者中脑脊液中S100B明显升高[(0.111±0.009)μg比(1.761±0.121)μg](P<0.05);与ANM患者发病期比较,经治疗恢复后脑脊液中S100B含量明显降低[(1.761±0.121)μg比(0.292±0.011)μg](P<0.05)。结论 S100B可作为评估ANM患者病情的重要标志物,其在发病期显著升高,症状改善后表达下调。
Objective To investigate the expression of S100B in cerebrospinal fluid of patients with acute non-specific myelitis(ANM) and the significance of S100B in assessing the condition and the status of the patient with ANM. Methods Eight patients with ANM admitted to the Affiliated Hospital of Logistics University of Chinese People's Armed Police Forces from Aug. 2015 to Aug. 2016 were enrolled in this study. The expression of S100B in cerebrospinal fluid was meas- ured by proteomics technique and verified by enzyme-linked immunosorbent assay (ELISA). S100B in healthy volunteers (N) and acute non-specific myalitis(S1) and after treatment(S2) were analyzed by ELISA to verify the accuracy of the results. Results For patients with ANM in acute phase, multiple high density shadows could be found by MRI examination; after treatment the symptoms alleviated, and the diffuse high density lesions disappeared. Proteomics S100B: SI: S2 = 0.33 ; S1 : N = 9.60 ; radioimmunoassay S100B : compared with healthy volunteers, the expression of S100B in cerebrospinal fluid ofANM patients in the acute phase was significantly increased[ (0.111± 0. 009 ) μg vs ( 1. 761 ± 0. 121 )μg ] ( P 〈 0.05 ). Compared with onset stage of patients with ANM, the content of S100B in cerebrospinal fluid was significantly decreased after treatment [ (1. 761 ±0.121 ) μg vs (0. 292 ±0. 011 ) μg ] (P 〈 0.05). Conclusion S100B can be used as an impor- tant marker for assessing the condition of patients with ANM,which is significantly increased at the onset stage and returns after treatment.
