摘要
以崇明水仙鳞片为外植体材料,对愈伤组织的诱导和再分化过程中的影响因素进行研究,摸索出最有利的组培条件,获得崇明水仙试管小鳞茎,并对组培过程中冷处理时间、鳞片位置及激素配比等影响因子进行研究。结果表明:鳞茎冷处理时间以3周为佳;内层鳞片诱导率和再分化率更高;冷处理3周的内层鳞片愈伤组织诱导和再分化的最佳培养基为MS+BA 1 mg/L+2,4-D 0.1 mg/L+蔗糖30 g/L+琼脂4 g/L,诱导率和再分化率分别为48.4%和23.3%。试验摸索出了鳞片愈伤组织途径获得崇明水仙试管小鳞茎的最佳条件,为崇明水仙的规模化生产提供技术支持。
In this study, the scales were used as explants to induce callus and organogenesis of Chongmingdaffodil, in order to obtain the most favorable conditions of tissue culture. The influencing factors of inductionof callus and organogenesis, such as low temperature treatment, the scales position and different combinationsand concentrations of hormones were analyzed. The results showed that: the inner scales treated with lowtemperature(4℃) for 3 weeks was the suitable explants for inducing callus, the induction rate was 48.4%. Thedifferentiation rate of callus was 23.3% on the MS medium with 1 mg/L BA, 0.1 mg/L 2,4-D, 30 g/L sucroseand 4 g/L agar. The experiment explored the optimal tissue culture condition to obtain the bulbs in vitro fromthe scales, and provided technical support for the large-scale production of Chongming daffodil.
