p27与cyclin D1在新疆哈萨克族食管鳞癌发生中的表达及临床意义

摘要 目的探讨p27与cyclin D1在新疆哈萨克族食管鳞癌中的表达及临床意义。方法应用逆转录-多聚酶链式反应(reverse transcription-polymerase chain reaction,RT-PCR)对48例新疆哈萨克族食管鳞癌组织及其配对的正常远端无癌组织进行p27与cyclin D1表达检测,并研究其表达与肿瘤分化、肿瘤分期[原发肿瘤,淋巴结,转移,(tumor,lymph node,metastasis,TNM)]、临床分期、淋巴结转移的关系。结果新疆哈萨克族食管鳞癌组织中p27与cyclin D1的阳性表达率分别为93.75%,93.75%,正常远端无癌组织的阳性表达率分别为97.92%,95.83%,二者之间差异无统计学意义(P=0.773,P=0.885,P>0.05)。在癌组织及正常远端无癌组织中p27在肿瘤分化、TNM分期、临床分期、淋巴结转移组织中的表达率分别为92.31%、100.00%,90.91%、94.59%,94.74%、93.10%,88.24%、96.77%。其表达与临床病理各因素间无相关性(均P>0.05)。cyclin D1在肿瘤分化、TNM分期、临床分期、淋巴结转移组织中的表达率分别为92.31%、100.00%,90.91%、94.59%,89.47%、96.55%,94.12%、93.55%。其表达与临床病理各因素间无相关性(均P>0.05)。结论 p27和cyclin D1的高表达伴随新疆哈萨克族食管鳞癌的发生,提示p27在新疆哈萨克族食管癌形成过程中未直接起负向调控作用。cyclin D1可能在新疆哈萨克族食管鳞癌早期发生中起正性调节作用,为食管癌发生的早期事件,有希望作为早期诊断指标之一。 [Objective] To explore the expression and clinical significance of p27 and cyclin D1 in Xinjiang Kazak's esophageal squamous cell carcinoma(ESCC). [Methods]The expression of p27 and cyclin D1 in 48 Xinjiang Kazak's ESCC tissue and matched normal distal non-cancerous tissue were detected by reverse transcription-polymerase chain reaction(RT-PCR). And the relationship of expression with tumor differentiation,tumor staging(TNM),clinical staging and lymph node metastasis were studied. [Results]The positive expression rates of p 27 and cyclin D1 in Xinjiang Kazak's ESCC tissue were 93.75% and93.75%,and in matched normal distal non-cancerous tissue were 97.92% and 95.83%,and the differences were not statistically significant(all P〈0.05). The expression rates of p27 in different tumor differentiation,TNM staging,clinical staging and lymph node metastasis in ESCC tissue and matched normal distal non-cancerous tissue were 92.31% 、100.00%,90.91% 、94.59%,94.74% 、93.10%,88.24% 、96.77% respectively. There are no correlation between the expression and clinical pathological factors(all P〈0.05). The expression rates of cyclin D1 in different tumor differentiation,TNM staging,clinical staging and lymph node metastasis were 92.31% 、100.00%,90.91% 、94.59%,89.47% 、96.55%,94.12% 、93.55% respectively. There are no correlation between the expression and clinical pathological factors(all P〉0.05). [Conclusion]The high expression of p27 and cyclin D1 perhaps participate in the development of ESCC. The results suggest that p 27 does not directly play a negative role in the regulation of Xinjiang kazak's ESCC formation. While cyclin D1 may play an important role in the early occurrence of ESCC,which is the early events of ESCC,and perhaps be an early diagnostic index.
