摘要
目的 :研究青蒿琥酯抗肝癌的作用机理。方法 :检测拓扑异构酶活性 ,甲基绿 派络宁染色检测凋亡细胞 ,免疫组化方法观察Bax、Bcl 2的表达。结果 :经青蒿琥酯处理的SMMC 772 1细胞拓扑异构酶活性增强 ,青蒿琥酯处理后 ,凋亡细胞明显增加 ,免疫组化显示 ,青蒿琥酯组肿瘤标本Bax蛋白阳性细胞数增多 ,Bcl 2蛋白阳性细胞数减少。结论 :青蒿琥酯抗肝癌作用机理可能为上调Bax基因 ,下调Bcl 2基因 ,诱导癌细胞凋亡 ,同时影响拓扑异构酶活性。
Objective:To study the mechanism of Artesunate inhibiting hepatocarcinoma.Methods:We assay the activity of the topoisomerase of hepatocarcinoma cells.The methyl green prynoin staining and the immunohistochemical staining was used to detect apoptotic cells and the tumor associated protein.Results:After the administration of Artesunate,the apopotic cells increased significantly,the immunohistochemical staining showed that Artesunate downregulates the expression of Bcl 2 genes and upregulates the expression of Bax genes.Conclusion:The mechanism of Artesunate may partly be inducing hepatocarcinoma cells to undergo apoptosis.Artesunate also interferes the activity of topoisomerase.
