采用植绒拭子二步法提取交通事故中的脱落细胞DNA——以一例交通肇事案为视角

摘要 在交通事故中肇事驾驶员为了逃避责任,会谎称自己不是驾驶员,此时驾驶员的确定对案件性质显得尤为重要。而在现场环境较为复杂的情况下,通常不易发现明显有价值的生物物证,因此对脱落细胞的发现和提取就具有非常重要的意义。在传统棉签二步转移法的基础上,通过两种不同方法对交通事故车辆上的生物检材进行提取,以及对两种提取方法 DNA检出进行差异分析。通过使用植绒拭子取代传统棉签,保留了传统棉签二步法:操作简单、不易污染、提取范围易于掌控的优点,使其适应性更强,使微量的脱落细胞更易捕获,获得了更高纯度的脱落细胞,提高了DNA的检出。 In a traffic accident, the driver denied that he is not the driver. However, it is the key to the case to identify whether he is the driver. In complex environment, it is often not easy to find valuable biological evidence, thus the discovery and extraction of exfoliated cells is of great significance. On the basis of the traditional two-step transfer method using cotton swabs, the biological samples on the traffic accident vehicles were extracted by two different methods, and the difference between the two methods was analyzed. Flocked swab is used instead of the traditional cotton swab, but still with the traditional two-step swab. Advantages of the method like simple operation, no pollution, easy to control making, more adaptable and the traces of the cells are easier to capture, higher purity is easier to achieve and DNA is easier to detect.
