摘要 建立一套完整的金花茶组培再生体系,包括无菌苗诱导、继代增殖、壮苗、生根、移栽等,且各阶段配方效果稳定.种子萌发诱导培养基:MS+6-BA 0.2 mg/L+NAA 0.2 mg/L+蔗糖3%的效果好,萌发率为82%;继代增殖培养基:改良MS+6-BA 1 mg/L+KT 1 mg/L+IBA 1.5 mg/L+蔗糖4%的有效苗最多,增殖系数为4.2;壮苗培养基以改良MS+6-BA 0.3 mg/L+IBA 1 mg/L+蔗糖3%的效果最好;生根培养:无菌苗基部浸泡500 mg/L IBA溶液2 min后接种于1/3改良MS+蔗糖1.5%培养基的生根率最高,达86%;移栽基质以V泥炭土∶V黄泥∶V河沙=2∶1∶1的混合基质较好,60 d后移栽成活率可达91%. A technique system of tissue culture of golden camellia was established completely in the study, including asepsis seedling induced, subculture multiplication, growth-promoting, rooting and transplantation. The techniques were as follows:asepsis seedling induced culture media was MS+6-BA0.2 mg/L+NAA0.2 mg/L+ Sucrose(3%), induction rate was 82%;clump shots induction culture media was modified MS +6-BA 1 mg/L+KT 1 mg/L+IBA 1 mg/L+ sucrose(4%), multiplication coefficients reached 4.2; growth-promoting culture media was modified MS+6-BA0.3 mg/L+IBA1 mg/L +sucrose(3%); rooting culture media was 1/3 modified MS +sucrose(1.5%), sterile shoots dipped in 500 mg/L IBA solution for 2 min were inoculated on the rooting culture media, and their rooting?rate was up to 86%; transplantation media was peaty soil 50% + yellow soil25%+fine sand 25%, and transplanting survival rate could reach 91% after 60 days.
