摘要
目的:检测遗传性角膜混浊突变系小鼠(B6-Co)与B6小鼠在胚胎后期眼睑组织中Hspa5、E-cadherin基因及蛋白的表达变化,探讨两者与B6-Co小鼠EOB表型形成的内在联系。方法:取B6、B6-Co胚胎期16.5天(E16.5d)、E18.5d小鼠眼睑,用Realtime PCR法和Western Blot法检测眼睑中Hspa5、E-cadherin的m RNA和蛋白表达情况。结果:E16.5d,B6-Co小鼠眼睑中Hspa5表达显著降低,E-cadherin表达显著升高;E18.5d,B6-Co小鼠Hspa5表达显著升高,E-cadherin表达显著降低。结论:Hspa5、E-cadherin的基因和蛋白在B6-Co突变系小鼠眼睑中的表达出现明显异常,提示B6-Co小鼠胚胎发育后期Hspa5、E-cadherin表达变化与EOB表型的形成有密切关联。
Objective:The expressions of both m RNA and protein of heat shock protein a5(Hspa5) and E-cadherin in the eyelid of B6-Co mice and B6 mice at the late embryonic stage were detected to investigate the internal relationship between the above two genes and the EOB(eye open at birth) phenotype of B6-Co mice.Methods:At embryonic day 16.5(E16.5 d) and E18.5 d,eyelids of B6 and B6-Co mice were taken to detect the expression of m RNA and protein of both Hspa5 and E-cadherin by Realtime PCR and Western Blot.Results:At E16.5 d,the expression of Hspa5 in the eyelids of B6-Co mice decreased significantly,and the expression of E-cadherin increased significantly; At E18.5 d,the expression of Hspa5 in the eyelids of B6-Co mice increased significantly,and the expression of E-cadherin decreased significantly.Conclusion:The expressions of m RNA and protein of both Hspa5 and E-cadherin in the eyelid of B6-Co mice were obvious abnormal,which revealed that the abnormalities of expressions of Hspa5 and E-cadherins at the late embryonic stage were closely associated with the EOB phenotype.
