摘要 [目的]建立组培固定番茄杂种优势优化体系。[方法]以番茄杂种优势组合优秀植株的侧芽作为外植体,MS培养基为基本培养基,通过添加不同浓度配比的6-BA和IAA筛选适宜的不定芽诱导、增殖培养基,并通过添加不同浓度的NAA筛选适宜的不定芽生根培养基。[结果]在温度25~26℃,光照强度3 000 lx,光照时间12 h/d的环境和静止培养的条件下,参试材料在MS+6-BA 2.4 mg/L+IAA 0.60 mg/L的培养基上能够诱导出生长状态良好的不定芽,并且能够大量增殖;在1/2MS培养基中添加的NAA浓度为0.5 mg/L时,不定芽生根状态较好。[结论]该研究建立的番茄杂种优势组合植株侧芽组培快繁体系,可以快速繁殖出大量番茄杂种优势组合侧芽的组培苗,将其用于固定番茄杂种优势,可以达到加快番茄新品种选育进程的目的。 [ Objective ] To establish optimization system of fixed tomato heterosis by plant tissue culture. [ Method ] Taking lateral bud from out-standing hybrid tomato plants as explant, MS culture medium as the base, to screen out the induction and enrichment medium for adventitious bud by addition of different ratios of 6-BA and IAA. Meanwhile, to screen out the rooting medium by adding different concentrations of NAA. [ Re-sult] By static culturing at the temperature of 25 -26 °C with illumination at 3 000 lx for 12 h/d,the testing explant can be inducted to a well growing adventitious bud in the culture medium containing MS + 6-BA 2.4 mg/L + IAA 0. 60 mg/L. This adventitious bud also can be massively enriched. Rooting status showed well in 1/2MS culture medium with NAA at the concentration of 0. 5 mg/L. [ Conclusion] The optimization sys-tem of fixed tomato heterosis by plant tissue culture can massively and quickly enrich the lateral bud of tomato with heterosis as tissue cultured seedlings, which can be used for fixing the heterosis of tomato and speeding up the cultivation of new tomato species.
