大鼠海马和杏仁核电点燃癫痫模型的比较 被引量:5

Comparison of epilepsy models induced by electrical stimulation in hippocampus and amygdala of rats

摘要 目的比较大鼠海马和杏仁核电刺激癫痫模型,探讨不同类型癫痫的病理机制。方法将健康成年雄性Sprague Dawley大鼠40只,随机分为海马癫痫模型组(EP-Hi组,n=16)、杏仁核癫痫模型组(EP-PAmy组,n=16)、假手术组(仅麻醉后切开头皮,插入电极但不做电刺激,n=8)。海马和杏仁核模型进行电刺激,观察大鼠的行为学表现;完全点燃之后2 h和35 d,将所有的实验大鼠用免疫组化法检测脑组织内c-Fos、NeuN的表达。结果行为学检测:杏仁核癫痫模型组(EPPAmy)的发作症状较海马癫痫模型组(EP-Hi)典型;免疫组化结果显示,与sham组相比,EP-PAmy组和EP-Hi组海马各区及皮质各区c-Fos阳性表达细胞数均明显增多(P<0.05),EP-Hi组海马各区c-Fos阳性表达细胞数明显高于EP-PAmy组(P<0.05),而皮质各区c-Fos阳性表达细胞数增加的程度较低(P<0.05);与sham组相比,EP-PAmy组和EP-Hi组海马及皮质各区神经元核蛋白(NeuN)阳性表达细胞数明显减少(P<0.05),EP-Hi组海马各区NeuN阳性表达细胞数明显低于EP-PAmy组(P<0.05),而皮质各区NeuN阳性表达细胞数减少的程度较低(P<0.05)。结论电刺激EP-PAmy的发作以及c-Fos、NeuN在海马各区和皮质各区的表达与EP-Hi是有差异的,从而表明EP-PAmy的神经环路对脑区的影响与EP-Hi不同。 Objective To compare the kindled epilepsy models of hippocampus and amygdala and to explore the pathological mechanism of different types of epilepsy. Methods Forty healthy adult male Sprague Dawley rats were randomly divided into epilepsy model of hippocampus group( EP-Hi group,n = 16),epilepsy model of amygdala group( EP-PAmy group,n = 16),sham group( n = 8). The hippocampal and amygdala epilepsy models were electrically stimulated to observe the behavioral performance of rats. At 2 h and 35 d after electrical kindling,all experimental rats were perfused with 4% paraformaldehyde and the brain tissues were taken. The expression of c-Fos and neuronal nucleoprotein( NeuN) was detected by immunohistochemical staining. Results The symptoms of the epilepsy model of periamygdaloid( EP-PAmy) were more typical than that of the epilepsy model of hippocampus( EP-Hi). Compared with sham group,the number of c-Fos positive cells by immunohistochemical staining were increased significantly in the hippocampus and cortex in EP-PAmy group and EP-Hi group( P 〈 0. 05). The number of c-Fos positive cells in hippocampus in EP-Hi group was significantly higher than that in EP-PAmy group( P 〈 0. 05),while the number of c-Fos positive cells in cortical regions in EP-Hi group was significantly lower than that in EP-PAmy group( P 〈 0. 05). Compared with sham group,the number of NeuN positive cells by immunohistochemical staining in the hippocampus and cortex were decreased significantly in EP-PAmy group and EP-Hi group( P 〈 0. 05). The number of NeuN positive cells in the hippocampus was significantly lower in EP-Hi group than in EP-PAmy group( P 〈 0. 05),while the number of NeuN positive cells in cortical regions was significantly higher in EP-Hi group than in EP-PAmy group( P 〈 0. 05).Conclusion The symptoms and the expression of c-Fos and NeuN in the hippocampus and cortex of EP-PAmy are different from those of EP-Hi,which suggests that the neural loop and the effect of EP-PAmy on the brain are different from those of EP-Hi.
