Real-time PCR方法检测早产儿脐带血中CMV-DNA、HSV1-DNA和HSV2-DNA的表达

The expression of CMV-DNA,HSV1-DNA and HSV2-DNA in umbilical cord blood of premature infants by real-time PCR

摘要 目的探讨早产儿脐带血中巨细胞病毒(CMV)、单纯疱疹病毒Ⅰ(HSV1),单纯疱疹病毒Ⅱ(HSV2)的感染与早产的相关性。方法采用实时荧光定量PCR(real-time PCR),对我院2015年6月1日-2016年12月31日出生的89例早产儿(研究组)和235例正常足月儿(对照组)脐带血进行检测。结果 89例早产儿中CMV-DNA检出10例,阳性率为11.24%,HSV1-DNA检出5例,阳性率为5.62%,HSV2-DNA检出4例,阳性率为4.49%。235例足月儿中CMV-DNA检出4例,阳性率为1.70%,HSV1-DNA检出2例,阳性率为0.85%,HSV2-DNA检出0例,阳性率为零。早产儿的CMV、HSV1、HSV2的感染率均高于足月儿,差异比较有统计学意义(P<0.05)。结论脐带血中CMV、HSV1、HSV2感染与早产的发生有明显相关性;且通过real-PCR方法检测较传统的检测方法敏感性强,准确度高。 Objective:To investigate the relationship between the infection of CMV,HSV1 and HSV2 in premature infant′s umbilical cord blood and premature delivery. Methods:Umbilical cord blood of 89 premature infants and 235 healthy term infants from June 1,2015 to December 31,2016 were detected by real-time PCR. Results:In 89 premature infants,10 cases(11.24%)were CMV-DNA positive,5 cases(5.62%)were HSV1-DNA positive,4 cases(4.49%)were HSV2-DNA positive. In 235 healthy term infants,4 cases(1.70%)were CMV-DNA positive,2 cases(0.85%)were HSV1-DNA positive,and all cases were HSV2-DNA negative. The infection rates of CMV,HSV1,HSV2 in premature infants were higher than those in healthy term infants,and the differences were statistical significant(P0.05). Conclusion:The infections of CMV,HSV1 and HSV2 can be early identified by real-time PCR. This method′s susceptibility and accuracy are stronger than traditional detection methods.
