CXC亚族趋化因子受体3及其配体I-TAC在直肠癌肝转移中的作用及临床意义

Expression and clinical significance of I-TAC and its receptor CXCR3 in rectal carcinoma with liver metastasis

目的 检测 CXC 亚族趋化因子受体 3（CXCR3） 及其配体 I-TAC 在直肠癌组织中的表达，并初步分析其在直肠癌肝转移中的作用及意义。方法 研究分组：研究组（30 例伴有肝转移的直肠癌组织标本）、对照组（46 例不伴有肝转移的直肠癌组织标本）。采用逆转录聚合酶链反应技术、免疫组织化学技术检测两组标本中 CXCR3 及 I-TAC 的表达水平。结果 研究组中 I-TAC 及 CXCR3 的 mRNA 和蛋白质表达水平均较对照组高。I-TAC 在研究组中阳性表达率（83.33%）高于对照组中阳性表达率（60.87%），两组比较差异有统计学意义（P ＝ 0.036）。CXCR3 在研究组中阳性表达率（70.00%）高于对照组中阳性表达率（54.35%），两组比较差异有统计学意义（P ＝ 0.021）。结论 伴肝转移直肠癌组织中存在 I-TAC及其受体 CXCR3 的特异性激活，提示 I-TAC/CXCR3 生物轴在直肠癌肝转移过程中发挥重要作用。

Objective To investigate the expression of I-TAC, CXCR3 in rectal cancer, and to reveal the role of I-TAC/CXCR3 in the liver metastasis of rectal carcinoma. Methods 30 rectal carcinoma tissue specimens with liver metastasis （research group） and 46 rectal carcinoma tissue specimens without liver metastasis （control group） were selected. The expression of I-TAC and CXCR3 in specimens were determined by reverse transcription-polymerase chain reaction and immunohistochemistry. Results The mRNA and protein expression of I-TAC and CXCR3 in the research group were significantly higher. The positive expression rate of I-TAC in the research group was significantly higher than that in the control group （83.33% vs.60.87%）, with statistically significant difference between the two groups （P=-0.036）. The positive expression rate of CXCR3 in the research group was significantly higher than that in the control group （70.00% vs.54.35%）, with statistically significant difference between the two groups （P=-0.021）. Conclusion Up-regulated expression of I-TAC and CXCR3 was associated with rectal cancer liver metastasis, I-TAC/CXCR3 signaling pathway may play a role in the process of rectal cancer liver metastasis.
