摘要
CD45分子是一种广泛表达在造血细胞表面跨膜蛋白酪氨酸磷酸酯酶(PTP ase),通过其胞质区PTPase活性,调节Src家族蛋白激酶,在淋巴细胞发育和活化中发挥关键作用,鉴于CD45分子在免疫细胞发育和活化中核心地位,已被选择作为免疫调节活性筛选一个重要靶点。综述CD45分子的结构、在不同淋巴细胞信号转导作用以及CD45分子与免疫调节剂开发最新研究进展,为进一步寻找新型调节剂提供思路。
CD45 is a family of transmembrane PTPs, which is exclusively expressed on the surface of all nucleated haematopoietic cells and their precursors except mature erythrocytes and platelets. The major intracellular target of CD45 PTPase activity is the Src -family kinase, which in T cells are predominantly p56LCK and p59Fyn (Fyn). LCK, in particular, is a primary initiator of T lymphocyte activation and development. Given that the cytoplasmic domain of CD45 contains PTPase activity and plays a role in TCR -mediated signaling, CIM5 was considered as important target for screening immunomodulatory activity. In order to provide the research idea on screening immunomodulator on CD45 PTPase screening assay, this paper reviewed the present progress on the structure of CD45 molecule, signal transduction in different lymphocyte and development of immunomodulator.
