附子理中汤对脾阳虚大鼠脂质过氧化损伤和能量代谢障碍的影响 被引量:19

摘要 目的:通过脾阳虚大鼠回肠组织中GSH-Px、MDA,线粒体呼吸链复合物Ⅰ、Ⅳ和Na^+-K^+-ATP酶、Ca^(2+)-Mg^(2+)-ATP酶的检测,及经附子理中汤干预治疗后三对指标变化情况,探讨脾阳虚证状态下大鼠脂质过氧化损伤及能量代谢障碍的机制,及附子理中汤改善脾阳虚证的作用机理。方法:采用经典复合因素造模方法塑造脾阳虚大鼠模型,将42只大鼠随机分为正常对照组、脾阳虚模型组、中药反证组,用ELISA检测大鼠回肠组织中GSH-Px、MDA,线粒体呼吸链复合物Ⅰ、Ⅳ和Na^+-K^+-ATP酶、Ca^(2+)-Mg^(2+)-ATP酶的表达水平。结果:脾阳虚状态下,大鼠回肠组织中GSH-Px、线粒体呼吸链复合物Ⅰ、Ⅳ和Na^+-K^+-ATP酶、Ca^(2+)-Mg^(2+)-ATP酶活性均显著降低(P<0.05,P<0.01),而MDA含量呈上升的趋势(P<0.01);经附子理中汤中药治疗后,其活性均有所回升,但仍低于正常组(P<0.05,P<0.01),MDA含量也有所下调(P<0.05)。结论:脾阳虚证状态下及中药治疗后,抗氧化酶与能量代谢相关酶的活性变化,脂质过氧化损伤及能量代谢障碍的影响,提示可能与脾阳虚证的病理机制及附子理中汤的作用机理有关。 Objective : spleen deficiency in rat ileum of GSH - Px, MDA, detection of mitochondrial respiratory chain complexes Ⅰ, Ⅳ and Na^+ - K^+ - ATP enzyme and Ca^2+ - Mg^2+ - ATP enzyme, and the changes of the three indexes of Fuzilizhong decoction after treatment, to explore the mechanism of spleen yang deficiency rats under the condition of lipid peroxidation damage and energy metabolism, and Fuzilizhong decoction canimprove the mechanism of spleen yang deficiency syndrome. Methods : using the classical modeling methods of complex factors shaping the spleen yang deficiency rat model, 42 rats were randomly divided into normal control group, model group, Chinese medicine disproof of spleen deficiency in rat ileum group, with ELISA to test GSH - Px and MDA detection, the expression level of mitochondrial respira- tory chain complex I , mitoehondrial respiratory chain complex IV, Na^+ - K^+ - ATP enzyme and Ca^2+ - Mg^2+ - ATP enzyme activity. Results : spleen yang deficiency syndrome, ileum tissue of rat GSH - Px, mitochondrial respiratory chain complex Ⅰ , mitochondrial respiratory chain complex IV, Na ^+ - K^+ - ATP enzyme and Ca^2+ - Mg^2+ - ATP enzyme activity were significantly decreased (P 〈 0. 05, P 〈 0.01 ), while MDA content was increased (P 〈 0. 01 ) ; the Fuzilizhong decoction, its activity has rebounded, but still lower than the normalgroup (P 〈0. 05, P 〈0. 01 ), the content of MDA content has also been reduced ( P 〈 0. 05 ). Conclusion : the syndrome of spleen yang deficiency and the treatment of traditional Chinese medicine, the activity changes of antioxidant enzymes associated with energy metabolism enzymes, lipid peroxidation damage and energy metabolism disorder, suggesting possible mechanism and pathological mechanism of spleen yang deficiency and Fuzilizhong Decoction on.
