针刺对乳腺增生大鼠乳腺组织Fasl、Fas表达的影响

Effects of Acupuncture on Fasl and Fas Expressions of Hyperplasia Rats' Breast Glands

目的:探究针刺对乳腺增生大鼠乳腺组织中Fasl和Fas蛋白表达的影响。方法:选取40只SPF级健康雌性未孕SD大鼠,12周龄,随机分为4组:正常组、模型组、针刺组、三苯氧胺组,每组10只。经25 d造模成功后,进行4周的干预治疗。HE染色法对乳腺组织进行病理学观察;免疫组化法、Western Blot法检测各组大鼠乳腺组织Fasl、Fas的蛋白表达情况。结果:与正常组相比,模型组乳腺小叶显著增多,腺泡结构紊乱,组织增生现象明显,Fas蛋白表达水平明显降低(P<0.01),Fasl蛋白表达水平明显增高(P<0.01);经治疗后,针刺组与三苯氧胺组较模型组小叶数量减少,腺泡排列较为整齐,组织增生的病理变化减轻,Fas蛋白表达水平明显增高(P<0.05或P<0.01),Fasl蛋白表达水平明显降低(P<0.05或P<0.01)。结论:针刺能够干预乳腺增生大鼠乳腺组织中Fasl和Fas蛋白表达情况,对乳腺增生大鼠有调节和治疗作用。

Objective ： To explore effects of acupuncture on Fasl and Fas protein expressions of experimental hyperplasia rats＇breast gland. Methods： We chose 40 female, 12 weeks aged and non- pregnant SD rats, and then randomly divided them into four groups： normal group, model group, acupuncture group, tamoxifen group. Each group had 10 rats. After the success of disease model with 25 days, treatment intervention was 4 weeks. HE staining was used to observe the pathological changes of breast tissue. The immunohistochemistry and Western blot were used to observe Fasl and Fas protein expressions of each group＇s breast gland. Result：Compared with the normal group, lobuli mammae increased significantly of the model group. The disorder of acinar structure and tissue hyperplasia phenomenon were obvious. The expression of Fas was significantly decreased （P 〈 0.01 ） and the expression of Fasl protein was significantly increased （P 〈 0. 01 ）. After treatment, when the acupuncture group and tamoxifen group compared with the model group, the number of lobuli mammae was reduced,acinar cells arranged more orderly. The pathological changes of tissue hyperplasia were reduced and the expressions of Fas protein were significantly increased （ P 〈 0. 05 or P 〈 0.01 ） and the expression of Fasl protein decreased significantly （P 〈 0.05 or P 〈 0.01 ）. Conclusion： Acupuncture can affect breast glands＇expressions of Fasl and Fas protein, regulate and treat hyperplasia of mammary glands in rats.
