油菜蜂花粉酚类物质的抗氧化能力比较 被引量:5

Comparison of antioxidant activity of phenolic compounds from rape bee pollen

摘要 通过对油菜蜂花粉中酚类物质的抗氧化活性进行评价和比较,得到高抗氧化活性的酚类物质组分,为进一步研究其对氧化应激导致肝损伤的保护筛选研究材料。采用乙醇和超声辅助提取油菜蜂花粉中酚类物质,通过AB-8型大孔树脂吸附结合不同比例乙醇/水洗脱分离得到5个洗脱组分,测定各组分总酚、总黄酮含量,并测定和分析其DPPH·清除能力、ABTS+清除能力、还原力和抗脂质过氧化等抗氧化能力。结果表明:油菜蜂花粉乙醇提取物经过AB-8型大孔树脂分离后,60%乙醇/水洗脱组分达到较好的酚类物质富集的效果,并具有较好的DPPH·和ABTS^+清除能力,半抑制浓度(IC_(50))值分别为9.47μg/m L和14.15μg/m L,40%乙醇/水洗脱组分的还原力和抗脂质过氧化能力(IC_(50) 9.6μg/m L)较强,说明40%和60%组分具有较高的抗氧化活性。研究结果为进一步研究油菜蜂花粉酚类物质对氧化应激诱导肝损伤的保护作用提供了实验依据。 The aim of this study was to evaluate and compare the antioxidant activities of phenolic compounds in rape bee pollen, and obtain phenolic fractions with high antioxidant activities, in order to screen materials for the further study on their protective effects against oxidative stress induced liver injury. The phenolic compounds in rape bee pollen were extracted by ethanol and ultrasonic, and separated into five fractions by AB-8 macroporous adsorption resin with different ratio of ethanol/water elution. The total phenolics and flavonoids, DPPH- scavenging activity, ABTS+ scavenging activity,reducing power and anti-lipid peroxidation activity of different fractions were determined. The results showed that after the ethanol extracts of rape bee pollen separated by AB-8 macroporous resin, 60% ethanol/water elution fraction reached better enrichment of phenolic compounds, and had better DPPH and ABTS+ scavenging activities. The IC50 values were 9.47 μg/mL and 14.15 μg/mL. The 40% ethanol/ water elution fraction showed better reducing power and anti-lipid peroxidation activities (IC50 9.6 μg/ mL). The results indicated the 40% and 60% fractions have strong antioxidant activities, which provide an experimental basis for further study on the protective effects of phenolic compounds from rape bee pollen against oxidative stress induced liver injury.
