摘要
目的观察低聚果糖(FOS)对小鼠肠道黏膜的影响,并探讨其对结肠炎的治疗作用及可能机制。方法用5%DSS诱导建立小鼠结肠炎模型,将30例小鼠随机分成正常组、模型组、FOS组。FOS组在模型建立后饮用配置好的5%FOS溶液。观察各组疾病活动指数(DAI),实验结束后观察结肠长度变化、组织学损伤评分、菌群计数统计以及IL-6、IL-33、TNF-α、RegⅢγ的表达。结果 FOS组DAI(P<0.000 1)及组织学损伤评分(P<0.000 1)明显低于模型组,模型组小鼠结肠质地变硬且黏膜皱褶消失(P<0.01),FOS组结肠致病菌与模型组相比显著减少(P<0.01),FOS组结肠IL-6(P<0.05)、IL-33(P<0.05)的表达量相比于模型组显著降低,TNF-α(P>0.05)、RegⅢγ(P>0.05)虽有降低但无统计学意义。结论低聚果糖能够改善结肠炎小鼠有益菌群的比例,降低促炎细胞因子IL-33及IL-6的水平,减轻肠道炎症反应。
To explore effect of fructo-oligosaccharide(FOS) on the intestinal mucosa of mice, 30 mice weredivided into control group, DSS group, and FOS group. The mice in groups DSS and FOS were fed with 5% DSS toinduce inflammatory bowel disease and then the mice in group FOS were fed with 5% FOS. Disease activity index(DAI) of mice was calculated; the expression of IL-6, IL-33, TNF-α and Reg Ⅲ γ in colon of each group wasmeasured at the end of experiment. The changes of colon length and histological damage score and bacteria weremeasured too. The mice in group FOS showed a lower DAI(P0.000 1) and histological damage score(P0.000 1)as compared with group DSS. The colon texture of mice were hardened and the wrinkles of colon were disappeared(P〈0.01) in group DSS. The mice in group FOS showed a less pathogenic bacteria as compared with group DSS(P〈0.01). The levels of IL-6(P〈0.05), IL-33(P〈0.05), TNF-α and Reg Ⅲγ in colon of group FOS were lower thanthose in DSS at the end of experiment, while the difference of TNF-α and Reg Ⅲγ levels between the two groupdemonstrated no statistical significance. Taken together, treatment with FOS can improve the ratio of beneficial bacteria and reduce the expression of IL-6 and IL-33,thus decrease colonic inflammatory response in DSS induced colitis.
